二咖啡酰奎宁酸对脑缺血后星形胶质细胞Porimin指标的影响观察

摘　　要：目的观察二咖啡酰奎宁酸对脑缺血后星形胶质细胞Porimin指标的影响。方法 32只雄性SD健康清洁级大鼠,分为空白组、对照组及药物组。应用大脑中动脉阻塞模型(MCAO)在对照组和药物组诱导脑缺血发作后,在对照组中予生理盐水,在药物组中予二咖啡酰奎宁酸。在MCAO后6h、24h分别进行行为学神经功能评分,HE染色、Cresyl Violet染色和GFAP、Porimin、DAPI免疫荧光染色。结果对照组和药物组中HE染色及Cresyl Violet均显示缺血中心区脑组织坏死,缺血半暗带区细胞水肿,可见肿胀的星形胶质细胞,对照组细胞损伤重于药物组。免疫荧光检测示药物组Porimin与GFAP、DAPI三标阳性细胞数目显著低于对照组(P=0.01)。结论二咖啡酰奎宁酸能降低脑缺血后星形细胞porimin指标的表达,提示二咖啡酰奎宁酸在脑缺血时对星形胶质细胞起一定积极的保护作用。Objective To study the effect of dicaffeoylquinic acid on astrocyte porimin expression in cerebral ische- mia. Method A total of 30 male SD rat were divided into three groups ： blank group, control group, drug group. Cerebral ischemia was induced in the control group and drug group by occluding the middle cerebral artery. Saline was given in con- trol group and dicaffeoylquinic acid was given in drug group. 6h and 24h after MCAO ,the behavior and neurological func- tion scores were evaluated, HE staining, Cresyl violet staining and immunofluorescenee staining for glial fibrillary acidic pro- tein （GFAP）, Porimin ,4＇6-diamidino-2-phenylindole （DAPI） were performed. Result Both in control group and drug group, cresyl violet staining and HE staining showed the necrotic brain tissue in the center of ischemic areas, cell edema and astroeyte swelling of astroeytes were found in isehemie penumbra. The cell damage was more severe in the control group than that in the drug group. Immunofluorescence staining found that in the ischemic penumbra, the number of Porimin- GFAP and DAPI-positive cells in drug group was significantly lower than that in control group. Conclusion Dicaf- feoylquinie acid could reduce the astroeyte porimin expression in cerebral ischemia, indicating that dicaffeoylquinic acid might play an active role on astrocyte in cerebral ischemia.
