摘 要:目的研究番茄红素对2型糖尿病大鼠的降糖调脂和抗氧化作用。方法将60只经腹腔注射链脲佐菌素(STZ)制备的2型糖尿病大鼠模型分为模型组、番茄红素低剂量组(5 mg/kg)、番茄红素中剂量组(10 mg/kg)、番茄红素高剂量(20 mg/kg)组和盐酸二甲双胍(25 mg/kg)组,每组12只。另取12只同龄大鼠作为正常对照组。各药物组分别灌胃相应药物,模型组和正常对照组灌胃生理盐水,均每天1次,疗程4周。分别于灌胃0,2,4周测定各组大鼠空腹血糖及胰岛素水平。灌胃4周后,测量各组大鼠体质量、血清总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、总抗氧化能力(T-AOC)和丙二醛(MDA)水平;测定肝脏组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)的活性。结果与正常对照组相比,模型组胰岛素水平、体质量及HDL-C、TAOC水平均显著降低(P<0.01或P<0.05),空腹血糖、TC、TG、LDL-C、MDA水平显著升高(P<0.01或P<0.05);肝组织中SOD、GSH-Px、CAT活性显著降低(P<0.01)。与模型组相比,番茄红素中剂量组和番茄红素高剂量组体质量显著增加(P<0.05或P<0.01),血清TC、TG、LDL-C、MDA含量显著降低(P<0.01或P<0.05);番茄红素高剂量组胰岛素水平、HDL-C、T-AOC水平和肝脏组织中SOD、GSH-Px、CAT活性均显著升高(P<0.05或P<0.01),空腹血糖水平显著降低(P<0.05或P<0.01)。番茄红素高剂量组体质量和肝组织中SOD和CAT活性较番茄红素低剂量组和番茄红素中剂量组显著升高(P<0.05或P<0.01),血清MDA含量显著降低(P<0.05或P<0.01);番茄红素高剂量组胰岛素水平及HDL-C、T-AOC水平和肝组织中GSH-Px活性较番茄红素低剂量组显著升高(P均<0.05),血糖水平及TC、TG、LDL-C含量显著降低(P<0.05或P<0.01)。结论番茄红素对2型糖尿病大鼠具有剂量依赖性的降低血糖、改善血脂并提高机体抗氧化能力的作用。Objective It is to investigate the effects of Lycopene(LP) on blood sugar, plasma lipid and antioxidant ability in type 2 diabetic rats. Methods 60 diabetic rat models made by intraperitoneal injecting STZ were randomly divided into five groups: model group, LP 5, 10 and 20 mg/kg treated groups and metformin hydrochloride 25 mg/kg treated group, each group had 12 animals. Another 12 sameged rats were selected as normal group. The drug groups were given respective drugs while the model group and normal group given normal saline by intragastric administration once a day for 4 weeks. At 0, 2, 4 weeks after the drug was given, the level of blood sugar and insulin were determined. 4 weeks later, the body weight was detected ; the content of TC, TG, LDL - C, HDL - C and MDA, T - AOC in serum was determined ; the activity of SOD, GSH - Px, CAT in hepatic tissue serum were determined. Results Compared with normal control group, body weight, the levels of insulin, HDL - C, T - TOC and the activities of SOD, GSH - Px, CAT were significantly lower while the levels of blood sugar, TC, TG, LDL - C and MDA were higher in model group( P 〈 0.01 or P 〈 0.05 ). Compared with model control group, the body weight increased significantly while the contents of TC, TG, LDL - C and MDA in serum decreased in moderate and high dosage of LP groups (P 〈 0.05 or P 〈0.01) ; the levels of insulin, HDL - C and T - AOC, the activity of SOD, GSH - Px, CAT in hepatic tissue were significantly increased while the level of FPG decreased in high dose of LP group( P 〈 0.05 or P 〈 0.01 ). The body weight and the activity of SOD and CAT in hepatic tissue of high dose LP group were significantly increased while the content of MDA in serum were decreased compared to those in the low and moderate LP groups (P 〈 0.05 or P 〈 0.01 ).
