放牧山羊LPL基因的组织表达谱分析

摘　　要：为弄清放牧山羊LPL mRNA的组织表达规律,以及为探索提高瘦肉率而不降低肌内脂肪含量的育种方法奠定理论基础,以放牧的性成熟黔东南小香羊、贵州白山羊、贵州黑山羊、黔北麻羊和南江黄羊为对象,应用实时荧光定量技术检测心、肝、肺、肾、背最长肌、半膜肌及皮下脂肪中LPL基因的表达.结果表明,放牧山羊LPL基因主要在心、背最长肌、半膜肌和皮下脂肪中表达,以心内丰度最高;肝和肾中也有表达,但水平较低;肺中偶有表达.结果提示,放牧山羊各组织中LPL的mRNA表达丰度不同,LPL对各组织脂肪含量的影响程度存在差异.In order to understand the tissue distribution regularities of LPL mRNA in grazing goats, and to establish the breeding theories for exploring the breeding methods of improving the lean meat rate and without reducing the intramuscular fat content, sexual maturity old of grazing goats including Qiandongnan small - xiang goats, Guizhou white goats, Guizhou black goats, Qianbei Ma goats and Nanjiang yellow goats were regarded as research objects, and the expression levels of LPL gene in liv- ers, kidneys, hearts, lungs, longissimus muscle, semimembranosus muscle and subcutaneous fat were detected by real - time fluorescence quantitative RT - PCR method. The results showed that LPL gene in grazing goat was mainly expressed in heart, longissimus muscle, semimembranosus muscle and sub- cutaneous fat, and with the highest abundance in heart. Furthermore, it was also expressed in liver and kidney, but the levels were low. And meanwhile, it was expressed in lung occasionally. The re- sults suggested that the abundances of LPL mRNA in tissues of grazing goats were different, and the effects of LPL on the fat content in different tissues were different, too.
