摘  要:以GR3木薯(Manihot esculenta Crantz)胚性愈伤组织为材料.研究了培养基中添加不同浓度蔗糖、甘露醇和烯效唑对不同保存时间木薯胚性愈伤组织SOD活性的影响。结果表明.处理组与对照组SOD活性呈先升高后降低的趋势,且在保存30d时SOD活性同时出现峰值:与对照相比,随着蔗糖浓度不断升高,SOD活性在蔗糖浓度25—35g/L胁迫下呈上升趋势,而高浓度(40~45g/L)蔗糖胁迫下呈下降趋势;随着甘露醇浓度的提高,SOD活性不断升高,且一直维持较高水平,浓度达到30g/L时开始呈现缓慢下降的趋势;随着烯效唑浓度的升高,SOD活性不断升高,烯效唑浓度为6mg/L时开始下降,浓度8~10mg/L时SOD活性下降速度较一致。蔗糖和甘露醇处理比烯效唑处理的SOD活性高.下降的速度也较缓慢,说明适当浓度的蔗糖和甘露醇,可以有效地提高木著胚性愈伤细胞活力.延缓细胞衰老,有利于木薯愈伤组织的继代保存.With material as cassava varieties GR3 embryonic callus, the SOD enzyme activity of different saving days cassava embryonic callus by adding different concentration of sucrose, mannitol and olefin to medium was studied. The results showed that the SOD enzyme activity was up before a decline in the treatment and control groups, and storing for 30 d, the SOD enzyme activity was in peak; Compared with the control group, with increasing concentration of sucrose, the SOD enzyme activity was rising by low concentration (25-35 g/L), and in the high concentration (40~45 g/L) was falling; with increasing concentration of mannitol, the SOD enzyme activity was rising and maintaining at a high level, until the concentration at 30 g/L, it began to slow decline; With olefine concentration increasing, the SOD enzyme activity was rising, when the concentration at 6 mg/L, it was decline, and same of decline rate between 8~10 mg/L. Compared with the olefine, the SOD en- zyme was high activity and decline slowly of sucrose and mannitol, the experiment showed that the suitable concentration of sucrose and mannitol can effectively improve the cell vitality and delay cell senescence, is helpful to preservation of cassava callus transgenerational.
