摘  要:目的:探讨蝎毒(scorpid poison)与眼镜蛇毒(cobra venom)联合应用对胃癌细胞BGC-803凋亡的影响及作用机制。方法:用蝎毒和(或)眼镜蛇毒处理体外传代培养的胃癌细胞株BGC-803。荧光显微镜下观察细胞增殖变化、进行活细胞计数及计算细胞增殖抑制率,流式细胞仪检测细胞凋亡,RT-PCR观察凋亡相关基因Bcl-2表达的变化。结果:蝎毒与眼镜蛇毒联合应用相对于单用蝎毒或眼镜蛇毒可显著提高胃癌细胞凋亡率、抗凋亡基因Bcl-2表达明显下降(P<0.05)。随着蝎毒和眼镜蛇毒合用浓度的增高,胃癌细胞凋亡率增加、抗凋亡基因Bcl-2表达下降更明显(P<0.05)。同一药物浓度作用24h与48h相比,亦有明显差异(P<0.05)。结论:蝎毒与眼镜蛇毒联合应用具有协同抗胃癌细胞的作用,并具有浓度和时间依赖性,其机制可能与增强诱导胃癌细胞凋亡及调控凋亡相关基因Bcl-2的表达有关。Objective:To study the anti-tumor effects of scorpid poison combination with cobra venom to BGC-803 of gastric cancer cell in vitro and to explore its possible mechanism. Methods:To treat cultured gastric cancer cell line BGC-803 with scorpid poison and( or)cobra venom. Viable cell was counted by trypan blue stain and cell pro-liferation inhibitive rate was calculated,cell apoptosis rate was determined by flow cytometry. Observed expression of apoptosis-related genes of Bcl-2 by RT-PCR. Results:Scorpid poison combined with cobra venom had a stronger inhibition function on BGC-803 of gastric cancer cell than scorpid poison or cobra venom,the expression of Bcl-2 was down(p﹤0. 05). As the concentrations increased of scorpion venom and cobra venom,difference was more sig-nificant(p﹤0. 05). In the same drug concentration effect,24 hours compared to 48 hours also statistically significant (p﹤0. 05). Conclusion:Scorpid poison combined with cobra venom have the synergistic effects on gastric cancer cell,and the mechanisms may be related to apoptosis-inducing effects and through regulating the expression of the related genes Bcl-2.
