摘  要:目的:研究Wnt信号通路激活剂R-spondin1在人骨髓间充质干细胞(h BMSC)成骨分化中的作用。方法:用0、5、10、20μg/L R-spondin1处理h BMSC,利用荧光素酶实验检测R-spondin1对h BMSC中Wnt信号通路的作用,Western blot检测R-spondin1对Wnt信号通路下游靶基因β-catenin蛋白表达的影响;在成骨分化培养基中添加20μg/L R-spondin1诱导h BMSC成骨分化,检测R-spondin1对早期成骨分化指标ALP活性及成骨分化晚期钙沉积、成骨分化标志物OSX、OCN和成骨分化关键转录因子RUNX2表达的影响。结果:R-spondin1增强h BMSC中Wnt信号通路,R-spondin1增强ALP活性和钙沉积,促进OSX、OCN及RUNX2的表达。结论:Rspondin1增强h BMSC中Wnt信号通路的作用,促进h BMSC成骨分化。Objective: To investigate the effect of R-spondin1,a Wnt signal pathway activator,on osteogenic differentiation of human bone marrow mesenchymal stem cells( hBMSC). Methods:hBMSC were treated with 0,5,10,20 μg / L R-spondin1. Luciferase assay was employed to detect the effect of R-spondin1 on Wnt signal pathway,and Western blot was used to detect the expression of β-catenin which was the downstream target gene of Wnt signal pathway. Alkaline phosphatase( ALP) activity,calcium deposition,and the levels of OSX,OCN,RUNX2 were detected to observe the effect of R-spondin1 on hBMSC osteogenic differentiation. Results: R-spondin1 enhanced Wnt signal pathway,ALP activity,calcium deposition and the expression of OSX,OCN and RUNX2 in hBMSC osteogenic differentiation. Conclusion: R-spondin1 can enhance Wnt signal pathway of hBMSC and promote osteogenic differentiation.
