摘  要:目的:探讨松树皮提取物原花青素对缺血再灌注小鼠脑损伤海马神经元的保护作用。方法:120只雄性昆明小鼠随机分成正常对照组、假手术组、14 d模型组、28 d模型组、14 d治疗组、28 d治疗组,采取小鼠双侧颈总动脉结扎缺血15 min,再灌注15 min构建脑缺血再灌注损伤模型,治疗组给予松树皮提取物原花青素灌胃治疗,Morris水迷宫实验观察小鼠学习记忆能力的变化,造模后第14、28天处死小鼠,HE染色法观察小鼠脑组织病理形态学改变,透射电子显微镜观察海马神经元超微结构的改变。结果:Morris水迷宫结果显示模型组小鼠学习记忆能力降低,与模型组相比,治疗组小鼠学习记忆能力有所提升;14 d模型组脑细胞数量较假手术组明显减少,胞浆着色浅,胞核移位,治疗组尼氏体排列欠规律,部分细胞形态不规则,胞浆着色相对较浅,但比模型组好;电镜下,小鼠海马CA3区神经元在14 d模型组损伤严重,神经元胞体水肿、出现核固缩溶解现象,存在细胞器溶解后形成的空泡,线粒体嵴可见有断裂,细胞出现凋亡小体,28 d模型组有所恢复,14 d和28 d治疗组神经元较同期模型组轻。结论:松树皮提取物原花青素对脑缺血再灌注损伤小鼠具有保护作用。Objective: To explore the protective effect of procyanidin extracted from pine bark on hippocampus neurons of mice subjected to ischemia reperfusion brain injury. Methods: A total of 120 male Kunming mice were randomly divided into 6 groups,namely normal control group,sham operation group,14-day model group,28-day model group,14-day treatment group and 28-day treatment group. The model of cerebral ischemia-reperfusion injury was constructed by bilateral carotid ligation ischemia for 15 minutes,and ischemia reperfusion for another 15 minutes. The treatment group was continuously given procyanidins by intragastric administration for 7 days before execution,and other groups were given the same amount of normal saline by intragastric administration. The change of memory capacity was observed in Morris water maze test. When mice were killed 14 and 28 days after model construction,the pathological changes of mice brain tissue were observed by HE staining method and the hippocampus ultrastructure was observed by transmission electron microscope. Results: The result of Morris water maze test showed that compared with model group,learning and memorizing ability in treatment group was improved to some extent. Compared with sham operation group,cells number in14-day model group was significantly decreased,cytoplasmic staining was light and cell nucleus was tanslocated. In treatment group the tigroid body was arranged irregularly,some cells was in irregular shape and cytoplasmic staining was relatively light. However,the situation in treatment group was better than that in model group. In 14-day model group,hippocampus CA3 neurons of mice were seriously damaged,pericaryon was in edema,karyopyknosis dissolving phenomenon occurred,organelles dissolved and formed vacuole,the mitochondrial cristae fracture was broken,and the apoptotic bodies occurred in cells.
