摘  要:目的检测唐氏综合征(Down syndrome,DS)脐带血与外周血单个核细胞中micro RNA差异表达图谱,进行比较分析。方法 Illumina检测脐带血与外周血中micro RNA的表达水平,比较两组间的差异表达,并进行靶基因预测与基因功能注释。结果脐带血与外周血显著差异表达的micro RNA分别有344和253种,分析可知,外周血表达上调的mi RNA增加了179种,下调的减少了270种。GO分析有5个生物学过程在预测的靶基因中被有意义的富集(P<0.001)。结论 DS脐带血与外周血micro RNA的表达差异显著,可能与胎儿生长发育过程中免疫系统缺陷,智力低下等症状的发生,发展相关。Objective: To detect the microRNA differentially expression profiles in mononuclear cells from Down syndrome umbilical cord blood and peripheral blood, and comparative analysis. Methods: Illumina was used to detect the microRNA differentially expression in cord blood and peripheral blood samples, and the target gene biological function was analyzed. Results: A total of 344 and 253 miRNAs were found significantly dysregulated expression in the DS cord blood and peripheral blood compared to the controls, respectively. A total of 179 and 270 miRNAs were found significantly expression in the DS peripheral blood mononuclear cells compared to the cord blood. A total of 5 GO (gene ontology) terms in biological processes were significantly enriched (P〈0.001) . Conclusion: These results suggested that there is a significant difference between the expression patterns of miRNAs in the DS peripheral blood group and cord blood group, which may be related to the immune deficiency, mental retardation development proceeding of DS fetus.
