摘 要:目的:建立湖北产葛根药材及提取物的指纹图谱检测方法,为确保葛根药材、提取物及其制剂的内在质量提供依据.方法:采用TIANHE Kormasil C18柱;流动相A:甲醇,流动相B:1%醋酸溶液;流速1.0mL·min-1;检测波长为250nm;柱温为35℃.梯度洗脱程序:0~10min 22%A,10~40min 22%~45%A,40~45min 45%A.对湖北省十堰竹溪、恩施州和黄冈罗田三个产地的葛根药材及提取物进行指纹图谱研究.结果:湖北省三个产地葛根药材及提取物的指纹图谱基本一致.结论:湖北三个产地的葛根药材质量差异不大.葛根药材、提取物相关性良好,各产地指纹图谱基本一致,制备工艺稳定可行.Objective: To establish the fingerprints of Radix Puerariae and its extract in Hubei Province for the quality control.Methods : The fingerprints of Radix Puerariae and its extract were established by HP LC. The chromatographic separation was per-fermed on a TIANHE Kormasil C18 column(4.6 mmX 150 mm, 5m). The mobile phase consisted of methanol and 1 % acetic acidwith gradient elution. The f low-rate was 1.0mL · min-1 , the column temperature was at 35℃and the UV absorbance detection wasat 250nm. Results: Under the selected chromate graphic conditions, a good fingerprint of Radix Puerariae and its extract were ob-tained. The fingerprint of Radix Puerariae from Zhuxi Shiyan, Enshi and Luotian Huanggang were basically identical. Conclusion:The quality of Radix Puerariae and its extract can be controlled effectively by HPLC-UV fingerprint. The Preparation process of theextract from Radix Puerariae is stable and feasible.
