摘  要:目的观察虎杖提取物白藜芦醇(Rev)对放射诱导小鼠肺损伤的保护作用及其防护机制。方法将30只C57BL/6雄性小鼠实验小鼠随机分为对照组、照射组和照射给药组,每组各10只。照射组和照射给药组小鼠用1%戊巴比妥钠(30 mg/kg)腹腔注射麻醉后固定于照射板架上,每日接受1次137Cs源γ射线胸部照射,共照射5次。对照组小鼠接受伪照射。照射给药组照射期间及照射后2周予Rev 40 mg/kg每日灌胃给药,对照组和照射组予等量4.8%乙醇载体灌胃。末次照射后15 d,全身麻醉,处死小鼠。观察3组小鼠肺组织病理改变,检测丙二醇(MDA)含量,超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GPX1)活性和基因表达水平。结果与照射组比较,照射给药组在小鼠肺泡炎水平明显改善,肺组织MDA含量显著减少,SOD及GPX1酶活性升高,SOD2及GPX1基因表达水平升高(P<0.05)。结论虎杖提取物Rev可以提高放射性肺损伤小鼠肺组织的氧化应激水平,改善肺组织炎症。Objective To observe the protective effects and protective mechanism of resveratrol (Rev) ex-tracted from Polygonum cuspidatum on radiation induced lung injury in mice .Methods 30 C57BL/6 male laborato-ry mice were randomly divided into control group , irradiation group and irradiation drug administration group , 10 mice of each group .Mice in irradiation group and irradiation drug administration group received peritoneal injection of 1%pentobarbital sodium (30 mg/kg), and then were fastened to irradiation grillage .Mice in irradiation group and irra-diation drug administration group received thoracic irradiation of 137 Cs source gamma rays once daily , 5 times totally. Mice in control group received pseudo irradiation .Mice in irradiation drug administration group received daily intra-gastric administration of Rev (40 mg/kg) during irradiation and 2 weeks after irradiation.Mice in other two groups received daily intragastric administration of carrier of 4.8%ethanol at the same dose .Mice in all groups were sacri-ficed 15 days after the final irradiation .The pathological changes of lung tissue , the content of propanediol ( MDA) , the enzyme activities of Chaoyang tiger dismutase ( SOD) and glutathione peroxidase ( GPX1 ) and the expression of gene in mice were observed in 3 groups .Results The level of pulmonary alveolitis in irradiation drug administration group had obviously improved as compared with that in irradiation group , and the content of MDA in lung tissue was decreased , the activities of SOD and GPX 1 were increased , the gene expressions of SOD 2 and GPX1 were increased ( P 〈0.05).Conclusion Resveratrol extracted from Polygonum cuspidatum can increase the level of oxidative stress in lung tissue of irradiating mice , and improve the infection reaction of lung tissue .
