摘  要:目的构建MCP-1的真核表达载体,并转染小鼠恶性黑色素瘤细胞株B16,从而构建含有趋化因子MCP-1的小鼠恶性黑色素瘤疫苗,研究该疫苗在体外对淋巴细胞的趋化作用。方法通过对目的基因的提取来构建重组质粒p EGFP-N1-MCP-1并转染裸鼠恶性黑色素瘤细胞株B16,观察其对单个核细胞体外趋化的活性。结果趋化型肿瘤细胞疫苗B16-MCP-1组细胞侵袭数目明显高于空白培养基阴性对照组及野生型B16疫苗组,而野生型B16疫苗组趋化的单个核细胞数目与阴性对照组相似。结论成功构建MCP-1真核表达载体p EGFP-N1-MCP-1及小鼠趋化型肿瘤疫苗B16-MCP-1,该疫苗在体外具有趋化小鼠单个核细胞的功能。Objective To construct murine malignant melanoma caccine and explore its chemiotaxis to lymhocytes in vitro, the eukaryotic expression vector of MCP-1 was constructed and murine malignant melanoma cell line B16 was transfected. Methods The target gene was extracted to recombine plasmid pEGFP-N1-MCP-1 and transfect malignant melanoma cell line B16 in the nude mice so as to observe its chemotactic activity to the mononuclear cells in vitro. Results The number of cells invaded in the chemotactic tumor cell vaccine B16-MCP-1 group was significantly larger than that in the negative control group and the wild type B16 vaccine group, while the number of chemotactic mononuclear cells in B16-MCP-1 group was similar to that in the negative control group. Conclusion Successfully constructed as mentioned above, the vaccine’s chemotactic function on the murine mononuclear cells is confirmed.
