摘  要:目的观察研究VEGF单克隆抗体对动物模型实体肿瘤生长的影响,为临床应用提供实验依据。方法选用BALB/c裸鼠10只,将细胞浓度为5×107个/ml的人舌癌细胞,分别用皮试针接种于裸鼠右前肢背部皮下,0.2ml/只,细胞数量为1×107个,注射完毕后随机分实验组和对照组,每组5只。对照组注射200μl Nacl,实验组注射200μl VEGF单克隆抗体,每天1次,持续28天,处死,进行大体观察和临床试验。结果实验组(VEGF单克隆处理组)移植瘤体积显著低于对照组(P<0.01);肿瘤最终重量低于对照组(P<0.05);实验后期,对照组的体重要明显高于实验组(P<0.05)。结论 VEGF单克隆抗体可显著抑制体内移植瘤的生长。Objective To investigate the influence of VEGF monoclonal antibody on the growth of the solid tumor of animai and provide experimental basis for the clinical application. Methods Ten BALB/e nude mice were involved in this study, and 0.2ml tongue squamous cell carcinoma cells (5 ×10^7 cells/ml) were inoculated into the subcutaneous of right forelimb back of each mouse. The nude mice were divided randomly into the experimental group and the control group, with 5 mice each group. Two-hundred of microlitre VEGF monoclonal antibody was injected into the surrounding of xenograft in experimental group while 200ul of normal saline was injected into the surrounding of xenograft in control group once a day for 28 days. Then these nude mice were sacrificed for general observation and clinic trials. Results The volume of xenografts in the experimental group (with VEGF monoclonal treatment) was lower than that of the control group (P〈0.01). Compared with the control group, the final weight of xenografts in the experimental group was lower (P〈0.05). In the late stage of the experiment, the weight of xenografts in the control group was distinctly higher than that of the experimental group (P〈0.05). Conclusion VEGF monoclonal antibody can significantly inhibit the growth of xenograft in vivo.
