摘  要:目的探讨蓝光暴露对视网膜色素上皮(Retinal Pigment Epithelial,RPE)细胞的形态和c AMP-PKA信号的通路影响。方法利用蓝光仪对RPE细胞进行0 min、20 min、40 min和60 min的蓝光照射。在光暴露后30 min、4 h和24 h,采用倒置相差显微镜观察RPE细胞的形态学变化;在光暴露后30 min,采用ELISA试剂盒检测细胞上清液c AMP和PKA浓度的变化。结果 RPE细胞密度随光暴露时间增加而逐渐降低,包膜包裹的细胞碎片随光暴露时间增加而逐渐增多,细胞形态由贴壁生长呈多边形向悬浮生长呈圆形或类圆形改变。在蓝光暴露后,c AMP浓度随光暴露时间延长而呈显著下降(P<0.05);PKA浓度在前40 min内并未显著下降,而在60 min时其浓度急剧下降。结论蓝光暴露可致视网膜色素上皮细胞中c AMP-PKA信号通路下调,诱导凋亡发生,且细胞损伤愈严重,c AMP和PKA浓度愈低。Objective This study aimed to in vitro investigate the influence of blue light exposure on the morphology and c AMP-PKA signal pathway in cultured human retinal pigment epithelium(RPE) cells. Methods After blue light exposure for 0min, 20 min, 40 min and 60 min, the morphology of RPE cells were observed by inverted phase contrast microscope. The concentrations of c AMP and PKA in the cellular supernatant were evaluated by c AMP and PKA ELISA kit. Results After blue light exposure RPE cell density gradually decreased, a loss of cell viability and an increase of apoptotic bodies were also observed. The concentration of c AMP in the supernatant significant decreased with exposure time(P〈0.05). PKA concentration did not change significantly within the 40 min after exposure, but declined sharply after a 60-min blue light exposure. Conclusion The blue light exposure could down-regulate c AMP-PKA signal pathway, leading to the development of cell apoptosis. With the development of apoptosis, the expression of c AMP-PKA pathway was further decreased by the damages in cells.
