摘 要:目的应用离子交联法制备色素上皮衍生因子壳聚糖(PEDF/CS)纳米粒,考察其体外性质及其在兔角膜碱烧伤中对角膜新生血管的影响,探讨其作用机制。方法通过离子交联法制备PEDF/CS纳米粒,考察纳米粒粒径、包封率、zeta电位及体外释放量,建立兔角膜碱烧伤模型,右眼作为实验眼。单纯随机抽样法,分为生理盐水对照组(A组)、PEDF滴眼液组(B组)、PEDF/CS纳米粒组(C组),每组20只。用药后7、14、21、28d大体观察及HE染色角膜新生血管情况,并使用RT-PCR方法检测各组角膜组织中VEGF的表达。结果 PEDF/CS纳米粒大部分呈规则的球型,粒径为(354±1.6)nm,zeta电位为(22.5±0.7)m V,包封率为(76.15±2.80)%,体系较为稳定,具有缓释功能。明显下调兔创伤角膜中的VEGF的表达促进角膜修复,各时间点各组间差异具有统计学意义(P<0.01)。结论色素上皮衍生因子壳聚糖纳米粒滴眼液通过下调兔创伤角膜中的VEGF的表达,可以减少角膜新生血管的生成。Objective To prepare the pigment epithelium-derived factor/chitosan (PEDF/CS) nanoparticles with Ionic crosslinking method, to investigate its properties in vitro and its influence on corneal neovascularization in rabbit cornea alkali burn, and to discuss its mechanism. Methods PEDF/CS nanoparticles were prepared with lonic crosslinking method, the nanoparticle size, encapsulation rate, zeta-potential and drug release in vitro were investigated; rabbit cornea alkali burn models were built in 60 New Zealand rabbits, the right eyes were chosen to be the experimental eye; the 60 rabbits were randomly divided into 3 groups: group A (saline control group), group B (PEDF eye drop group) and group C ( PEDF/CS nanoparticles group), 20 rabbits in each; at the time points of 7, 14, 21 and 28 days after drug giving, corneal neovascularization was observed by HE staining and VEGF expression in cornea tissue in each group was detected by RT-PCR. Results Most PEDF/CS nanoparticles were in uniform sphere with the particle diameter of 354±1.6nm, zeta-potential of 22.5±0.7mV and encapsulation rate of (76.15±2.80)%, the system was relatively stable with sustained-releasing function; VEGF expression was obviously down-regulated, which promoted the corneal tissue reparation; the differences among groups at each time point were of statistical significance (P〈0.01). Conclusions PEDF/CS nanoparticle eye drop can reduce corneal neovascularization by down-regulating VEGF expression in rabbit injured cornea.
