摘 要:目的研究细胞因子IL-27在急性肺损伤中发挥的作用和相关机制。方法采用盲肠结扎穿孔术(CLP)建立小鼠急性肺损伤模型。ELISA和荧光实时定量PCR法检测CLP-急性肺损伤小鼠模型的肺组织、支气管灌洗液、外周血单个核细胞及血清中IL-27及其亚单位的表达。同时,小鼠行CLP术前吸入给药IL-27中和抗体,经组织学检测小鼠肺组织炎症水平的改变,并检测支气管灌洗液中IL-6、CXCL10、IFN-γ的水平和小鼠存活率。结果 CLP-小鼠的肺组织和外周单个核细胞中IL-27的2个亚单位(EBI3和p28)均增加,支气管灌洗液和血清中的IL-27水平也较对照组显著增加(P<0.01)。抗IL-27多克隆抗体的使用降低了CLP-急性肺损伤小鼠肺组织的炎症水平,支气管灌洗液中IL-6、CXCL10、IFN-γ水平下降(P<0.05),同时提高了小鼠存活率。结论IL-27在急性肺损伤中发挥重要作用,抑制IL-27的表达可能成为预防和治疗急性肺损伤的新途径。This study designed to investigate the roles of IL-27 in mouse acute lung injury and the intrinsic mechanisms. A mouse acute lung injury model was established by cecal ligation and puncture(CLP) method. Then enzyme-linked immunosorbent assay and real-time fluorescence quantitative PCR assay were applied to detect IL-27 and its subunits expression in lung tissue, bronchoalveolar lavage fluid, and peripheral blood monocytes. On the other hand, IL-27 polyclonal antibody was used to treat mice before CLP, and then the inflammation level of lung tissue was evaluated by histological examination, the levels of inflammation factors in bronchoalveolar lavage fluid and the survival rate of mice were detected. Data showed that both EBI3 and P28 were increased in mouse lung tissue and peripheral monocytes after CLP. IL-27 in the bronchoalveolar lavage fluid and serum also increased significantly compared with the control group(P〈0.01). The using of IL-27 polyclonal antibody could not only reduce the inflammation levels in lung tissue after CLP, and reduce the levels of IL-6, CXCL10, IFN-γ in bronchoalveolar lavage fluid(P〈0.05), but also improve the mouse survival rate. Taken together, IL-27 plays an important role in acute lung injury, thus inhibiting the expression of IL-27 may be a new way for the prevention and treatment of acute lung injury.
