摘 要:目的检测mi R-21-5p、mi R-155-5p、mi R-218-5p、mi R-222-3p、mi R-494-3p和HMGB1在乳腺癌荷瘤小鼠肿瘤微环境中的表达水平,分析它们在乳腺癌发生、发展中的作用。方法收集乳腺癌MCF-7荷瘤小鼠手术标本;实时荧光定量PCR检测18只乳腺癌荷瘤小鼠手术标本中mi R-21-5p、mi R-155-5p、mi R-218-5p、mi R-222-3p、mi R-494-3p和HMGB1的m RNA的表达水平;Western blot法检测HMGB1在癌组织及癌旁组织中蛋白表达情况。结果与癌旁组织相比,肿瘤组织中mi R-21-5p、mi R-222-3p表达上调(P<0.05),mi R-155-5p、mi R-218-5p、mi R-494-3p表达下调(P<0.05);与癌旁组织相比,HMGB1在癌组织中高表达(P<0.05)。相关性分析发现,mi R-21-5p、mi R-222-3p与HMGB1的表达成正相关,而mi R-155-5p、mi R-218-5p、mi R-494-3p与HMGB1的表达成负相关。结论肿瘤微环境中mi R-21-5p、mi R-155-5p、mi R-218-5p、mi R-222-3p、mi R-494-3p和HMGB1可能在乳腺癌的发生、发展中起一定作用。This study designed to investigate the levels of mi R-21-5p, mi R-155-5p, mi R-218-5p, mi R-222-3p, mi R-494-3p and HMGB1 in tumor microenvironment of breast cancer and explore their clinical significance and correlations. Firstly, the surgical specimens of tumor-bearing mice were collect. RT-q PCR was employed to measure the m RNAs expression of mi R-21-5p, mi R-155-5p, mi R-218-5p, mi R-222-3p, mi R-494-3p and HMGB1 in tumor tissues and tumor-adjacent normal tissues from 18 tumor-bearing mice; Western blot was used to detect HMGB1 protein expression in tumor tissues and tumor-adjacent normal tissues from tumor-bearing mice.Data showed that comparing with the normal tissues, tumor tissues were characterized by over-expression of mi R-21-5p, mi R-222-3p(P〈0.05), and by down-expression of mi R-155-5p, mi R-218-5p and mi R-494-3p(P〈0.05).Additionally, tumor tissues were characterized by over-expression of HMGB1(P〈0.05). The over-expression of mi R-21-5p, mi R-222-3p and the up-regulation of HMGB1 were in positive correlation, but mi R-155-5p, mi R-218-5p and mi R-494-3p negatively correlated with HMGB1 in tumor microenvironment of breast cancer. In conclusion, all the result suggested that mi R-21-5p, mi R-155-5p, mi R-218-5p, mi R-222-3p, mi R-494-3p might play important roles in the carcinogenic process of breast cancer.
