摘 要:目的:观察亚低温治疗对癫痫持续状态(SE)大鼠海马HSP27表达的影响,以探讨亚低温治疗的神经保护机制。方法:采用侧脑室内注射红藻氨酸(KA)建立SE模型,108只雄性SD大鼠随机均分为正常对照组(N组)、假手术组(F组)、SE模型组(SE组)、亚低温治疗组(MT组),根据大鼠处死时间再分为6,12和24h共3个亚组。采用HE染色观察海马组织神经细胞变化,用免疫组化法检测海马组织中HSP27的表达。结果:MT组大鼠癫痫发作级别及神经元损害较SE组降低;SE组3个亚组HSP27表达较F组相对应的各亚组明显增多(P<0.05);MT组中3个亚组HSP27的表达较SE组相对应的各亚组明显增多(P<0.05);F组与N组比较,观察指标无统计学意义。同一观察组内6,12和24h,3个亚组HSP27表达呈上升趋势。结论:亚低温治疗对大鼠癫痫持续状态有神经保护作用,其神经保护的机制与上调HSP27表达有关。Objective: To observe the interventional effects of mild hypothermia treatment on expres sion of heat shock protein 27 (HSP27) in kainic acid (KA)-induced status epilepticus (SE) rats, along with the possible neuroprotective mechanism of mild hypothermia treatment. Methods: An animal model of status epilepticus was established in rats by intracerebroventricular injection of KA. 108 SD rats were randomly divided into normal control group (group N), sham operation group (group F), SE model group (group SE), and mild hypothermia treatment group (group MT). Each group was further divided into 3 subgroups according to execution time (6 h, 12 h and 24 h). Morphologic changes of the h expressions of HSP27 in hippocamp lp al pocampal neurons were observed by HE staining, while the tissue measured by immunohistochemical techniques. Re- suits: Compared with group SE, seizure levels and neuronal damage decreased significantly in group MT, but HSP27 expressions increased in the three subgroups (P〈0.05). However, in hipp0cam- pal tissue of group SE, HSP27 expressions of the three subgroups were lower than those in group F (P〈0.05). And there was no statistical difference in HSP27 expressions between group N and g h roup F. In the same group, HSP27 expressions displayed an upward tendency from 6 h, 12 to 24 in 3 subgroups. Conclusion: Mild hypothermia treatment has protective effects on nerves of SE rats, which are related to the up-regulation of HSP27 expression.
