摘 要:目的:观察重症急性胰腺炎(SPA)对大鼠胃肠动力和肠神经系统回肠肌间神经节血管活性肠肽(VIP)和一氧化氮合酶(NOS)阳性神经元的影响。方法:雄性大鼠(SD)20只随机分为假手术组和SAP组,用5%牛黄胆酸钠逆行胰胆管穿刺注射制作SAP模型。24h后,胃内灌服1%台盼蓝溶液,测定小肠推进比。应用双重免疫荧光染色分析肌间神经节VIP和NOS阳性神经元。结果:SAP组胰腺病理评分增高,大鼠小肠推进比率(30.20±3.82)%,低于假手术组(51.37±5.04)%,差异有统计学意义(P<0.05);SAP组肌间神经节VIP和NOS阳性神经元,高于假手术组,差异有统计学意义(P<0.05)。结论:重症急性胰腺大鼠胃肠动力障碍可能与抑制性神经元VIP和NOS的表达上调相关。Objective:To investigate the effects of severe acute pancreatitis (SPA) on gastrointestinal motility and ganglia ileum muscle vasoactive intestinal peptide (VIP) and nitric oxide synthase (NOS) positive neurons in rats. Methods: 20 Sprague Dawley(SD) rats were randomly divided into two groups: sham operated group and SAP group. SAP model was established by retrograde injection of 5% sodium taurocholate into the bile-pancreatic duct. 24 h after the injection, stomach was filled with 1% trypan blue, then small intestine propulsion ratio was measured. Use double immunofluorescence to analyze myenteric ganglia VIP and NOS immunoreactive neurons. Results: In the SAP group, the histological score of the pancreas was increased, small intestine propulsion ratio (30.20±3.82) was lower than the sham operated group(51.37±5.04 )% ,the difference was statistically significant(P〈0.05);the myenteric ganglia VIP and NOS immunoreactive neurons percentage in the SAP group was higher than the sham operated group(P〈0.05). Conclusion: Gastrointestinal motility disorder of severe acute pancreatitis rat may be correlated with the upregulated expression of VIP and NOS in inhibitory neurons.
