摘  要:目的:观察全身垂直振动对去卵巢骨质疏松大鼠腰椎转录因子Osterix蛋白表达的影响,探讨全身垂直振动治疗绝经后骨质疏松的作用机制。方法:将36只3月龄健康雌性未孕SD大鼠随机分为假手术组(Sham组)、去卵巢静止组(OVX组)和去卵巢振动组(WBV组),每组12只。手术后10周,WBV组开始接受全身垂直振动治疗。振动频率90 Hz,重力加速度3.8 g,振幅0.5 mm,每天2次,每次15 min,每周7天,振动7周。振动结束后,腹主动脉取血处死各大鼠,按解剖位置分离第2腰椎(L2)和第3腰椎(L3)。用HE染色观察L2组织形态学变化;用TRAP染色法对L2石蜡切片破骨细胞进行染色并计数;用免疫组化检测L2转录因子Osterix定位变化;用Western blot检测L3转录因子Osterix蛋白表达水平的变化。结果:与Sham组和WBV组比较,OVX组大鼠腰椎骨髓腔脂肪空泡数目、骨小梁表面破骨细胞数目显著增加,而转录因子Osterix蛋白表达显著下降。结论:全身垂直振动能降低去卵巢骨质疏松大鼠腰椎破骨细胞的数目,增强腰椎转录因子Osterix蛋白的表达。Objective This paper investigated the effects of the whole-body vertical vibration(WBV) on the protein expression of transcription factor osterix in lumbar of ovariectomized osteoporosis rats in order to explore the therapeutic mechanism of the WBV for treating osteoporosis. Methods Thirty six 3-month old female healthy non-pregnant SD rats were randomly divided into sham-operation group(Sham, n =12)ovariectomized group(OVX, n=12) and ovariectomized plus whole-body vertical vibration group(WBV, n=12). After 10 weeks of ovariectomy, rats in WBV group were treated with 15-minute WBV(vibration frequency = 90 Hz, gravity acceleration = 3.8g) twice a day,7 days per week. After 7 weeks of WBV treatment, blood was drawn from abdominal aorta and second lumbar(L2) and the third lumbar(L3) were separated. The number of osteoclasts in L2 was calculated and the protein expression of osterix in L3 was detected. Results The number of bone marrow adipocytes and osteoclasts in OVX group was significantly higher than that in the other two groups, while the expression of L3 transcription factor osterix in OVX group was significantly lower than that in the other two groups. Conclusion The whole-body vertical vibration decreases the number of osteoclasts and increases the protein expression of the transcription factor osterix in OVX osteoporosis rats.
