摘  要:目的探讨5-HTTLPR、5-HT2A受体基因(5-HTR2A)T102C多态性和多巴胺β羟化酶基因(DβH)5'侧翼区-1021C/T多态性对注意缺陷多动障碍儿童行为问题的影响,为儿童基因学科的基础研究提供理论数据。方法应用PCR-限制性片段长度多态性(PCR-RFLP)方法检测包头市某小学974名小学生的5-HTTLPR、5-HTR2A的T102C多态性及DβH5'侧翼区-1021C/T多态性,采用t检验分析3种基因多态性对ADHD相关行为问题的影响。结果在冲动多动的得分上,5-HTTLPR的LL基因型的得分高于SS/SL基因型(P=0.007),在焦虑的得分上SS/SL基因型的得分高于LL基因型的得分(P=0.010);在品行问题、焦虑、多动指数的得分上,5-HTR2A的TC/CC基因型均高于TT基因型(P值均<0.05);DβH5'侧翼区的CC与CT/TT基因型在行为问题的得分差异均无统计学意义(P值均>0.05)。结论遗传因素可能影响ADHD儿童的行为问题表现。Objective To investigate the association of 5- HTTLPR,5- HT2 A receptor polymorphism and DβH-1021 C / T polymorphism with attention deficit hyperactivity disorder in children. Methods Polymerase Chain reaction( PCR) and RFLP technique were used to test 5- HTTLPR,5- HT2 A receptor T102 C and DβH- 1021 C / T polymorphism. Association of polymorphism with ADHD was analyzed. Results The scores of impulse hyperactivity in LL genotype was higher than SS / SL genotype(P = 0. 007). Score of anxiety in SS/SL genotype was higher than LL genotype(P = 0. 010). Conduct problem,anxiety,hyperactivity index in TC / CC genotype was higher than TT genotype( P〈 0. 05). There were no differences in behavioral problems in CC and CT / TT of DβH- 1021 C / T. Conclusion Genetic factors may affect the behavioral problems of ADHD. Conduct problem,anxiety,hyperactivity index in TC / CC genotype of 5- HTR2 A is higher than TT genotype of 5- HTR2 A.
