摘  要:目的 探讨碳水化合物反应元件结合蛋白(ChREBP)、脂肪酸合酶(FASN)与膀胱移行上皮癌(BTCC)临床病理特征之间的关系,分析其与微小RNA(miRNA,miR)-107、Snail1的表达相关性.方法 选用膀胱移行细胞癌手术切除标本48例,每例标本均选用癌组织、癌旁组织以及远端正常黏膜作为对照.采用反转录-聚合酶链反应(RT-PCR)检测切除标本中的miR-107、Snail1、ChREBP、FASN的表达,并分析与各临床病理特征之间的关系.结果 (1)ChREBP、FASN在BTCC组织中的表达明显高于正常黏膜组织(12.278±4.035比8.442±7.658,13.536 ±4.776比9.183±6.112,P<0.05),其在高级别尿路上皮癌中的表达高于乳头状瘤,与肿瘤分化程度有关(14.963±5.367比10.738±5.332,14.558±7.728比9.738±5.332,P<0.05),与肿瘤浸润深度、淋巴结转移无明显相关(P>0.05).(2) miR-107与ChREBP、FASN在BTCC中表达呈正相关,癌旁组织中miR-107、Snail1与ChREBP、FASN表达呈负相关(67.5%比32.4%,73.0%比27.0%,P<0.05).结论 ChREBP、FASN与膀胱移行上皮癌病理分级相关,miR-107可能通过调控ChREBP、FASN通路,改变肿瘤细胞内传统代谢通路,促进膀胱癌进展.Objective To investigate the expression level of carbohydrate response element binding protein (ChREBP) and fatty acid synthase (FASN) in the different clinical stages of bladder transitional cell carcinoma (BTCC),and explore the relationship between microRNA (miRNA,miR)-107 and Snail1 pathway with clinicopathological features of BTCC.Methods Forty-eight cases of BTCC specimens surgically resected were selected.For each specimen,the cancerous tissue,peri-cancer and its remote normal mucosa were obtained.The expression of miR-107,Snail1,ChREBP,and FASN in resected cancer tissues was detected by reverse transcriptase-polymerase chain reaction (RT-PCR),and its relationship with clinicopathologic features of BTCC was analyzed.Results (1) The expression rate of ChREBP and FASN was significantly higher in BTCC tissues than in normal tissues (12.278 ± 4.035 vs.8.442 ± 7.658,and 13.536 ± 4.776 vs.9.183 ± 6.112,P < 0.05).The expression of miR-107 in high grade urothelial carcinoma group was significantly higher than that in papillary epithelioma group,and miR-107 was related with the degree of tumor differentiation (14.963 ± 5.367 vs.10.738 ± 5.332,and 14.558 ±7.728 vs.9.738 ±5.332,P <0.05),but not with tumor depth of invasion and lymph node metastasis (P > 0.05).miR-107 was positively related to ChREBP and FASN expression in BTCC tissues,and miR-107 and Snail1 expression was inversely correlated to ChREBP and FASN in peri-cancer tissues (67.5% vs.32.4%,and73.0% vs.27.0%,P<0.05).Conclusion ThemiR-107 was related with the pathological grading of BTCC,which may change the traditional metabolic pathway to mediate the occurrence and development of bladder cancer through the regulation of ChREBP and FASN.
