摘  要:目的 检测Med-19基因在肾细胞癌组织中的表达水平,探讨肾细胞癌的生物学特征与其表达之间的关系.方法 应用免疫组织化学、反转录-聚合酶链反应(RT-PCR)及Western blot技术检测60例肾细胞癌组织、20例正常肾脏组织中Med-19基因的表达,并分析其与临床特征之间的关系.结果 免疫组织化学分析显示Med-19在正常肾脏组织中表达率为30.0% (6/20),而肾癌组织中Med-19的表达率为76.6% (46/60),提示Med-19基因在肾癌组织中显著上调(P<0.05);Western blot检测到Med-19蛋白表达的共48例,表达水平为0.618±0.125;但Med-19蛋白表达仅在2例正常肾脏标本中被检测到,其表达水平为0.076±0.023,差异有统计学意义(t=18.13,P<0.05);Med-19 mRNA的表达显示肿瘤标本中,50例检测到Med-19 mRNA的表达,其表达水平为0.863 ±0.081;Med-19 mRNA的表达仅在1例正常肾脏组织中被检测到,其表达水平为0.102±0.039,两者差异有统计学意义(t=18.98,P<0.05).肾细胞癌组织中Med-19基因mRNA和蛋白的表达水平显著高于正常肾脏组织(P<0.05);其表达水平与肿瘤的直径、临床分期、分化程度、有无淋巴结转移等生物学行为显著相关(P<0.05);与性别、年龄无明显相关(P>0.05);Spearman等级相关分析显示Med-19基因mRNA与蛋白表达呈正相关(r=0.776,P<0.05).结论 Med-19基因在肾细胞癌中过度表达,有助于判断肾细胞癌恶性转变以及肾细胞癌发生浸润转移.Objective To detect the expression of Med-19 gene mRNA and protein in renal cell carcinoma and to investigate the correlation between Med-19 gene expression and clinicalopathological features.Methods Med-19 gene was detected in 60 cases of renal cell carcinoma and 20 cases of normal renal tissues by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blotting.The correlation between the Med-19 expression and clinicopathological features was analyzed.Results Immunohistochemical analysis showed that the expression rate of Med-19 in renal cell carcinomas was increased significantly [76.6% (46/60),P < 0.05] as compared with that in normal renal tissues [30.0% (6/20)].Western blotting showed that the Med-19 protein was detected in a total of 48 renal cell carcinomas with the expression level being 0.618 ± 0.125,and that was detected in only 2 cases of normal renal specimens with the expression level being 0.076 ± 0.023 (t =18.13,P < 0.05).Med-19 mRNA expression was detected in 50 renal cell carcinomas with the expression level being 0.863 ± 0.081,and that was detected in only 1 normal renal tissue with the expression level being 0.102 ± 0.039 (t =18.98,P < 0.05).Med-19 gene mRNA and protein expression levels in renal cell carcinoma tissues were significantly higher than those in normal renal tissues (P < 0.05).There were significant correlations between the expression level and the biological behaviours such as the diameter of the tumor,clinical stage,differentiation,biological behaviors,and lymph node metastasis (P < 0.05),but no correlations between the expression level with gender and age (P > 0.05).Spearman rank correlation analysis indicated that the expression of Med-19 gene mRNA was positively correlated with its protein (P < 0.05).Conclusion Overexpression of Med-19 gene in renal cell carcinoma may be associated with tumor progression.
