摘　　要：目的 观察乙醇、微小RNA（miRNA）-219-5p对肝细胞肝癌（HCC）中聚糖蛋白3（GPC3）表达的影响.方法 采用免疫组织化学检测30例正常肝脏组织、120例HCC患者肝癌组织及相应癌旁组织标本中GPC3蛋白的表达水平,分析GPC3与HCC汉族临床资料之间的关系,探讨其与饮酒、miRNA-219-5p相关性.结果 GPC3在HCC组织中的阳性表达率为75％,在癌旁组织中其阳性表达率为30％,正常肝组织中未发现其阳性表达,3组比较差异有统计学意义（x2=74.348,P＜0.01）.在年龄、乙型肝炎表面抗原（HBsAg）阳性、慢性肝病病史以及肿瘤大小方面,GPC3表达均未受到影响[x2（肝癌组织）=1.800、0.533、1.140、0.568,P＞0.05;x2（癌旁组织）=0.179、0、0、0.714,P＞0.05];在大量饮酒的患者中,GPC3阳性表达率为100％,在非大量饮酒的患者中,其阳性表达率为70％,两组间差异有统计学意义（x2=8.000,P＜0.01）,提示饮酒行为对GPC3的表达明显相关.结论 HCC的发生与miRNA-219-5p表达过低、GPC3蛋白表达过高有关,同时大量饮酒行为可能放大了此种效应.Objective To observe the effects of alcohol and microRNA （miRNA）-219-5p on proteoglycan 3 （GPC3） in hepatocullular carcinoma （HCC）.Methods Immunohistochemical staining method was used to detect the expression of 30 cases of normal liver tissues,120 cases of HCC tissues and corresponding para-concerous liver tissues.The relationship between GPC3 and clinical data of Han patients with HCC,and the inner link between GPC3 with alcohol and miRNA-219-5p were analyzed.Results The GPC3 positive expression was 75％ in HCC tissues and 30％ in para-cancerous tissues （x2 =74.348,P ＜0.01）.No GPC3 expression was detected in normal liver tissues.The GPC3 expression was not related to age,positive hepatitis B surface antigen （HBsAg）,history of chronic liver disease and tumor size [x2（HCC tissues） =1.800,0.533,1.140,0.568,P ＞ 0.05;x2（para-cancerous tissues） =0.179,0,0,0.714,P ＞ 0.05）.The GPC3 positive expression rate was 100％ in patients with heavy drinking,and that was 70％ in non-heavy drinking patients.（x2 =8.000,P ＜ 0.01）,suggesting drinking behavior had a significant relevance with the GPC3 expression.Conclusion The occurrence of HCC is correlated with the low expression of miRAN-219 and high expression of GPC3 protein.At the same time,heavy drinking behavior can enlarge this effect.
