摘 要:目的 观察肉豆蔻挥发油对大鼠抑郁状态的影响并探讨其作用机制.方法 雄性大鼠72只,随机分为空白组、模型组、阳性对照组及肉豆蔻挥发油高、中、低剂量组,每组12只.除空白组外的其余组动物接受21 d不可预知刺激后,灌胃给药21 d,每天灌胃1h后给予不可预知的刺激.在造模后和给药后分别测定大鼠的水平运动、垂直运动和糖水消耗情况.行为学实验结束后分取大鼠全脑测定5-羟色胺、多巴胺和去甲肾上腺素递质含量及单胺氧化酶的活性.结果 慢性不可预知应激模型建立后,大鼠的水平活动及垂直活动次数、糖水偏好度均明显降低;给药后与模型组相比,肉豆蔻挥发油高剂量组表现出抗抑郁作用,水平运动[(33.22±4.21)分]及垂直运动[(9.56±1.74)分]情况明显改善、糖水偏好度增加(60.81±14.48)%,差异有统计学意义(P<0.01),其作用效果与盐酸氟西汀相比差异无统计学意义(P>0.05).肉豆蔻挥发油高剂量能显著提高大鼠脑内5-HT[(12.23±0.95) ng/ml]、DA[(179.11± 15.20) pg/ml]的含量以并降低单胺氧化酶的活性[(9.12±0.95) μg/ml],与模型组比较,差异具有统计学意义(P<0.01).结论 肉豆蔻挥发油对慢性不可预知应激模型大鼠具有明显的抗抑郁作用,其抗抑郁机制与提高大鼠脑内5-HT、DA的含量、降低脑内单胺氧化酶的活性有关.Objective To observe antidepressant effect of the volatile oil of Myristica on rats and investigate the mechanisms of antidepressant effect.Methods 72 rats were assigned randomly into normal control group,model group,positive control group and groups of different doses of the volatile oil of Myristica.After all rats except normal control group were applied to unpredictable stimulation and different doses of drugs for 21 days,and unpredictable stimulation after 1 h of gavaging.Horizontal and vertical activities and sugar consumptions were tested after molding and oral administration.After the behavioral experiments,the brains of the rats were taken,and 5-hydroxy tryptamine(5-HT),dopamine,noradrenalin content and activity of monoamine oxidase were measured.Results Comparing with model group after gavaging,the high dose of the volatile oil of Myristica showed antidepressant effect in the rats,the situation of crossing(33.22±4.21) and rearing(9.56± 1.74) was improved and sweet water consumption ((60.81± 14.48) %) of rats was increased,significantly comparison with model group (P< 0.01),but no significant difference comparison with FH(P>0.05).The high dose group could significantly increase the content of 5-HT((12.23±0.950) ng/ml),DA((179.11± 15.20) pg/ml) and reduce the activity of monoamine oxidase((9.12±0.95) μg/ml) in rats' brain,and significant differences were showed compared with model group(P< 0.01).Conclusion The volatile oil of Myristica has obvious antidepressant effect in rat chronic mild stress,its mechanism is related to increase the content of 5-HT and DA and reduce the activity of monoamine oxidase in rats ' brain.
