摘 要:目的 研究环氧化酶-2(COX-2)选择性抑制剂塞来考昔对癫癎持续状态大鼠脑组织中P-糖蛋白(P-gp)表达的影响,为寻找治疗难治性癫癎的新药物提供理论依据.方法 健康成年雄性SD大鼠60只按随机数字表法随机分为空白对照组、癫癎模型组和塞来考昔干预组.氯化锂-毛果芸香碱诱发SD大鼠癫癎持续状态,48只纳入实验,每组16只.各实验组均采用免疫组织化学法和Western blot法检测鼠额叶皮质区及海马区P-gp的表达情况.结果 免疫组织化学结果显示:与空白对照组比较,癫癎模型组中P-gp的表达显著增高,差异有统计学意义(P<0.01);塞来考昔干预后的大鼠脑组织P-gp较癫癎模型组显著下降,差异有统计学意义(P<0.01).Western blot法检测结果提示:各组额叶皮层和海马中均有P-gp的表达.与空白对照组比较,癫癎模型组中P-gp的表达量显著增高,差异有统计学意义(P<0.01);塞来考昔干预组大鼠脑组织P-gp的表达量较癫癎模型组显著下降,差异有统计学意义(P<0.05).结论 COX-2抑制剂塞来考昔可降低P-gp在癫癎持续状态大鼠脑组织中的表达,有望成为治疗难治性癫癎的新方法.Objective To study the effect of cyclooxygenase-2 (COX-2)selective inhibitor Celecoxib on the expression of P-glycoprotein(P-gp)in the brain of rats with status epilepficus,in order to assess the therapeutic value of intractable epilepsy.Methods Sixty adult male SD rats were randomly divided into blank control group,the epilepsy model group and Celecoxib intervention group.The status epilepticus was induced in rats by injecting Lithium pilocarpine.Forty-eight rats were included in the experiment.There were 16 rats in each of the blank control group,epilepsy model group and Celecoxib intervention group,respectively.Immunohistochemical method and Western blot method were used to detect the expression of P-gp in experimental group in the frontal cortex and hippocampus.Results Immunohistochemistry result showed that the expression of P-gp was significantly higher in epilepsy model group than the blank control group,and the difference was statistically significant (P 〈 0.01);The P-gp expression in the Celecoxib intervention group was lower than that in the epilepsy model group,and the difference was statistically significant (P 〈0.01).Western blot results suggested that the expression of P-gp could be found both in the frontal cortex and hippocampus in each group.Compared with the blank control group,the P-gp expression was significantly higher than that in the epilepsy model group,and the expression of the P-gp was lower after the Celecoxib intervention than that in the epilepsy model group,and the difference was statistically significant (P 〈 0.05).Conclusions COX-2 inhibitor Celecoxib could decrease the expression of P-gp in brain tissue with status epilepticus,which may provide a new method for the treatment of intractable epilepsy.
