摘 要:目的:研究姬松茸多糖(ABP-AW1)对OVA(ovalbumin,OVA)抗原诱导的细胞因子特异性转录因子的表达,探讨影响免疫应答的相关信号通路。方法:ABP-AW1和OVA皮下2点免疫ICR小鼠,于1 d,15 d各免疫1次,d 28分离培养脾细胞,免疫鼠脾淋巴细胞体外给予抗原OVA再刺激24 h。RT-PCR法分析ABP-AW1对脾细胞因子IFN-γ,IL-4mRNA表达水平及核转录因子T-bet和GATA-3mRNA的相对表达量,ELISA法检测培养上清细胞因子IFN-γ和IL-4的分泌。结果:ABP-AW1能增强OVA诱导的小鼠脾脏Tbet/GATA-3转录因子和Th特征性细胞因子基因表达以及脾细胞分泌水平的升高。结论:ABP-AW1通过IFN-γ/T-bet信号通路,调节OVA抗原的免疫应答机制。Objective: To investigate the effect of ABP-AW1 on expression of transcription factors of OVA immunized mice, so as to explore the signal pathway of the immune response. Methods: ICR mice were subcutane- ously immunized twice with OVA mixed in ABP-AW1 on day 1 and day 15. The splenocytes were acquired on day 28 and incubated in vitro with the same antigen for 24 h for re-stimulation. The production of cytokines IFN-y and IL-4 in cell culture supernate was detected by ELISA, and the mRNA expressions of cytokines mRNA and tran- scription factors T-bet and GATA-3 were detected by RT-PCR. Results: ABP-AW1 significantly enhanced the ex- pression of nuclear transcription factor T-bet and GATA-3. The levels of cytokine IL-4 and IFN-y were markedly in- creased also. Conclusion: ABP-AW1 can regulate the immune response through IFN-y/T-bet signal pathway.
