摘 要:目的:研究单体阿魏酸、绿原酸对血管平滑肌细胞(vascular smooth muscle cells,VSMCs)黏附的影响,并探讨相关机制。方法:体外培养VSMCs,用单体阿魏酸、绿原酸进行干预,检测其对VSMCs与基质的黏附、VSMCs之间的黏附的影响;PT-PCR检测对基质金属蛋白酶(matrix metal loproleinase,MMP)MMP-2、MMP-9 m RNA表达的影响。结果:(1)阿魏酸(102ng/m L、103ng/m L)、绿原酸(102ng/m L、103ng/m L)对VSMCs与基质的黏附无影响;(2)阿魏酸(102ng/m L、103ng/m L)促进了VSMC之间的黏附;(3)阿魏酸(102ng/m L、103ng/m L)可下调VSMCs MMP-9 m RNA的表达。结论:阿魏酸可促进VSMC之间的黏附,这种作用是通过抑制VSMCs MMP-9 m RNA的表达实现的。Objective : To investigate the effects of ferulic acid and eholorogenic acid on the adhesion of vascular smooth muscle cells (VSMCs) and its correlated mechanisms. Methods : VSMCs were in vitro cultured. The VSMCs adhesion was detected after in- tervened by ferulic acid and cholorogenic acid. The effects on the mRNA expressions of matrix metalloproteinase - 2/- 9 were de- tected using RT- PCR. Results:( 1 )Ferulic acid (102 ng/mL, 103 ng/mL)and cholorogenic acid( 102 ng/mL, 103 ng/mL)had no effects on VSMCs adhesion to ECM. (2) Ferulic acid ( 102 ng/mL, 103 ng/mL) promoted VSMCs and VSMCs adhesion. (3) Feru- lic acid (102 ng/mL, 103 ng/mL)could down- regulating MMP-9 mRNA expression. Conclusion :Ferulic acid promoted VSMCs and VSMCs adhesion through down -regulating MMP- 9 mRNA expression.
