摘  要:为解决目前工业用α-半乳糖苷酶耗能大、成本高的难题,研究泡菜汁中筛选出产α-半乳糖苷酶的菌株,而后对其进行紫外诱变和亚硝酸钠进一步诱变,并在不同温度和不同p H下测定α-半乳糖苷酶酶活力。结果表明:获得一株酶活高达36.9 U/m L的α-半乳糖苷酶菌株,该菌株所产α-半乳糖苷酶的最适反应温度为50℃,最适p H为5.5。利用4%海藻酸钠做为固定剂对α-半乳糖苷酶进行固定化,可以使酶活保持在70%左右,此α-半乳糖苷酶高产菌株的发现及其酶学特性的研究为α-半乳糖苷酶的工业化应用奠定了基础。In order to solve the high energy consumption,high cost problems of the industrial use of alpha galactosidase,we conducted the following experiments. An α-galactosidase strain was screened from pickle juice in this study. Ultraviolet mutation screening and sodium nitrite mutation were then conducted. The enzymatic activities of α-galactosidase enzyme were determined in different temperature and different p H. The results showed that α-galactosidase activity was increased to 36. 9U / m L through different mutation methods. Studies also showed that the optimal p H and temperature was 5. 5 and 50℃ respectively. 4% of sodium alginate which could remain α-galactosidase activity at about 70% was used as the fixing agent. It lay a foundation for the development of alpha galactosidase application.
