摘  要:目的探讨b FGF(外源性碱性成纤维细胞生长因子)基因转染骨髓间充质细胞(MSC)的鉴定方法。方法提取大鼠骨髓间充质细胞,体外培养后,随机分为b FGF组(构建b FGF基因病毒质粒,移植到MSC),正常组和阴性组(转染Ad.GFP),通过免疫組化染色法观察MSC的形态,同时应用Western-Blot法分析b FGF基因表达情况。结果阴性组和正常组均无转染b FGF,b FGF组胞浆呈黄色,Western-Blot分析可见b FGF特异性条带,其表达量为0.85,高于正常组及阴性组,P>0.05,差异不具有统计学意义。结论将病毒作为载体能够成功实现b FGF基因转染MSC,得到目的基因阳性表达。Objective To explore the identification method of basic fibroblast growth factor (bFGF) exogenous transfection to MSC (bone marrow cells). Methods The bone marrow mesenchymal cell of rats were extracted in vitro culture, divided into bFGF group (build bFGF gene plasmid and transplanted to MSC), normal group and negative group (transfection Ad.GFP) randomly. The MSC form was observed by immunohistochemical staining method, at the same time, analyzed bFGF gene expression by Western-Blot method. Results There were no transfected bFGF in negative group and normal group, bFGF cytoplasm was yellow, Western-Blot analysis showed the bFGF specific band, its expression quantity was 0.85, it was higher than t the normal control group and negative control group, P〉0.05 ,had no difference statistically significance. Conclusions The bFGF transfects to MSC gene can be successful by use virus as a vector, the positive expression can be obtained.
