摘  要:海陆蛙(Fejervarya cancrivora)是红树林半咸水生境中唯一的两栖动物,为了解铜离子(Cu2+)对该蝌蚪的毒性效应,测定了Cu2+对海陆蛙蝌蚪(38-39期)96 h内半数致死浓度LC50;在此基础上,模拟其野外生存环境,分别设计3个铜暴露组(CuSO4含量分别为80、160、300μg L-1)和对照组,检测海陆蛙蝌蚪在Cu2+暴露24 h、48 h、72 h、96 h后抗氧化能力、乙酰胆碱酯酶(AChE)活性及金属硫蛋白(MT)表达量的变化。结果显示,海陆蛙蝌蚪对Cu2+24 h、48 h、72 h、96 h LC50分别为5.69、3.37、1.34、0.95 mg L-1。在急性胁迫试验中,各暴露组蝌蚪体内超氧化物歧化酶(SOD)、ACh E活性以及MT表达均被抑制,在同一暴露时间下,处理组SOD、AChE活性和MT含量显著低于对照组;而在Cu2+胁迫下,蝌蚪体内脂质过氧化反应加剧,表现为丙二醛(MDA)含量显著增多,且染毒浓度越大、暴露时间越长,MDA含量越多。除AChE外,其他指标组染毒时间与浓度间具有显著的互作效应。本研究表明海陆蛙可作为红树林生境质量评价的指示物种,其中SOD、AChE活性及MDA含量可作为重金属Cu2+毒性效应评估较敏感的评价指标。The crab eating frog (Fejervarya cancrivora) is the only amphibian in the mangrove forest. To explore the feasibility of using its tadpole as an indicator of the habitat quality of mangrove forests, we studied the effect of acute copper exposure on the activities of superoxide dismutase (SOD) and acetylcholine esterase (ACHE) and the content of malonaldehyde (MDA) and metallothionein (MT) in the tadpole (Stage 38-39). The LC50 of Cu^2+ was 5.69 mg LI for 24 h exposure, 3.37 mg L1 for 48 h, 1.34 mg L1 for 72 h, and 0.95 mg L1 for 96 h exposure. Based on the 96h LC50, four treatments of Cu^2+ (control, 80, 160, 300μg·L^-1 CuSO4) were designed and sampled at 24 h, 48 h, 72 h and 96 h. The results showed that SOD and AChE activities and MT content in three treatment groups were lower than the control groups, and decreased significantly with the time and the increase of Cu^2+ concentration. However, the trend was conversed in the content of MDA. Higher exposure concentration and longer exposure time led to higher MDA content, suggesting that the lipid peroxidation increased under copper stress. Also, there was an interaction effect of exposure time and Cu^2+ content on all indicators except ACHE. Therefore, the biomarkers of SOD and AChE activity and MDA content are sensitive to heavy metal induced oxidative stress, and Fejervarya cancrivora can be the indicator species for environment monitoring of mangrove forests.
