摘 要:目的通过改良的糖氧剥夺(oxygen glucose deprivation,OGD)模型,探讨缺血缺氧中心区星形胶质细胞损伤后的细胞死亡途径与细胞膜损伤中钠钾ATP酶(Na+-K+-ATPase)活性及钠-钾-氯共转运体1(Na+-K+-2Cl--cotransporter 1,NKCC1)表达的关系。方法取出生24h内的SD大鼠海马星形胶质细胞进行体外培养,鉴定其纯度,应用改良的OGD模型,将星形胶质细胞进行OGD(0、1、2、3、4、6h)处理后检测不同时间点细胞的胀亡率、凋亡率、ATP含量、钠钾ATP酶的活性及NKCC1的表达水平。结果星形胶质细胞存活率随OGD时间的延长而降低,OGD 3h后胀亡细胞率超过凋亡细胞率。ATP含量和钠钾ATP酶活性随OGD时间的延长而下降,NKCC1的表达随着OGD时间的延长先增加后减少,在OGD 3h后明显减少。结论缺血缺氧中心区星形胶质细胞死亡在OGD 3h内以凋亡为主,OGD3h后胀亡是细胞死亡的主要方式,这种转化或许与Na+,K+-ATPase活性及NKCC1表达的激活有关。Objective To study the relationship between the astrocyte death pathway in the ischemic core and the activity of Na+,K+-ATPase or the expression of Na+-K+-2Cl--cotransporter 1(NKCC1)by using modified oxygen and glucose deprivation(OGD)model.Methods Astrocytes of the newborn SD rats(within 24h)were cultured in vitro and the cell purity identified.The oncosis rate and apoptosis rate of astrocytes,the content of ATP,the activity of Na+-K+-ATPase and the expression of NKCC1 were detected by use of the modified OGD model after OGD for 0,1,2,3,4or 6h.Results The viability of astrocytes was decreased with the extension of OGD.The oncosis rate was greater than the apoptosis rate after 3hOGD.The content of ATP and the activity of Na+-K+-ATPase were decreased with the extension of OGD.The expression of NKCC1 was first increased and then decreased with the extension of OGD,and it was significantly reduced after OGD for 3h.Conclusion The main death pathway of astrocytes in the ischemic core is apoptosis within 3hof OGD and becomes oncosis after 3-h OGD,which may be related to the Na+-K+-ATPase activity and the NKCC1 expression.
