摘  要:目的探讨口服抗生素是否引起大鼠结肠炎症以及合成去甲肾上腺素(Norepinephrine,NE)的酶即多巴胺β羟化酶(Dopamine-β-hydroxylase,Dβh)在结肠中的变化。方法在雄性SD大鼠的饮用水中添加肠道不吸收的抗生素(新霉素、杆菌肽和游霉素),饮用1周、3周之后分别取结肠组织进行组织病理学观察,并用实时定量PCR检测大鼠结肠促炎因子白细胞介素-1β(Interleukin-1β,IL-1β)和抗炎因子白细胞介素-10(Interleukin-10,IL-10)以及Dβh mRNA的表达水平。结果与对照组相比,饮用抗生素1周大鼠结肠HE(Haematoxylin and eosin)染色显示轻微的炎症,3周时结肠炎症略有加重;饮用抗生素1周、3周大鼠结肠IL-1β的mRNA水平上升(P<0.01),IL-10的mRNA水平下降(P<0.01),而Dβh的mRNA表达在1周时,与对照组比较差异无统计学意义,3周时显著升高(P<0.05)。结论大鼠口服抗生素可引起其结肠的轻度炎症并且结肠Dβh的表达在3周时显著升高。Objective To determine whether oral antibiotics could induce colon inflammation and influence the ex- pression of dopamine-β-hydroxylase (Dβh) in rats. Methods Male SD rats were given a mixture of nonabsorbable antimicrobials (ATM, containing neomycin, bacitracin and natamycin) via drinking water for 7 or 21 days respec- tively. Pathological changes were observed and the expressions of IL-1 β, IL-10 and Dβh in the colon were assessed by real-time PCR. Results Compared with the control group, the HE staining showed mild inflammation in one- week ATM treatment group, which was slightly severer after ATM treatment for three weeks. The IL-1 β levels were increased (P 〈0. 01 ) and IL-10 levels decreased (P 〈0. 01 ) after ATM treatment for both one and three weeks. Significant difference in the expression of Dβh was not observed in the one-week group but in the three-week group (P 〈 0.05). Conclusion Oral antibiotics can cause mild inflammation in rat colon, and the expression of Dβh in colon can significantly increase at three weeks.
