摘 要:目的:研究高盐、甜菜碱对同型半胱氨酸代谢的影响并探讨其机制。方法:50只昆明小鼠随机分为对照组(0.5%Na Cl饲料)、高盐组(8%Na Cl饲料)和干预组(8%Na Cl饲料+2%betaine饮水)喂养8周,比较各组血清渗透压、Hcy、Glu指标数值变化。H4IIE细胞分为空白对照组、高盐组(50 mmol/L Na Cl)、干预组(50 mmol/L Na Cl+1%甜菜碱)和干预对照组(1%甜菜碱),细胞密度达80%时收集细胞,Western blot检测BHMT表达量。结果:与对照组相比,高盐组血清渗透压、Hcy、Glu显著升高(P<0.05),BHMT表达量显著下降(P<0.05),干预组无差异(P>0.05);与高盐组相比,干预组血清渗透压、Hcy、Glu显著降低(P<0.05),BHMT表达量显著升高(P<0.05)。结论:高盐胁迫降低BHMT表达量,升高血清Hcy、Glu水平,甜菜碱能显著改善高盐胁迫对Hcy代谢的影响。Objective: To investigate the effect of high salt and betaine on homocysteine metabolism and explore its mechanism. Methods: 50 KM mice were randomly divided into control group (0.5% NaCI diet), high salt group (8% NaCI diet) and intervention group (8% NaCI diet and 2% betaine water). 8 weeks after treatment, levels of the serum osmolality, Hcy, Glu in each group are detected compared. H411E cells are divided into blank control group, high salt group (50 mmol/L NaCI), intervention group (50 mmol/ L NaCl+1% betiane) and intervention control group (1% betaine), cells were harvested when the density of cells reached 80% and the expression of BHMT were tested by Western blot. Results: Compared to control group, the levels of serum osmolality, Hcy, Glu in high salt group increased significantly (P〈0.05), the expression of BHMT reduced significantly (P〈0.05), no difference were detected comparing with intervention group (P〉0.05). Compared to high salt group, the levels of serum osmolality, Hcy, Glu in intervention group decreased (P〈0.05), the expression of BHMT increased (P〈0.05). Conclusion: High salt stress decreased the expression of BHMT, thus resulting into the increase of serum Hcy level. Betaine can considerably ameliorate the effect of high salt stress on homocysteine metabolism.
