摘 要:目的以果蝇为对象研究山楂提取物(hawthorn fruit extract,HFE)延长寿命和抗氧化的作用。方法收集2d龄的雄性和雌性果蝇,平均分为4组,饲喂添加不同浓度(0、0.5、1.5、4.5mg/ml)山楂提取物的培养基。研究HFE对果蝇生存寿命,超氧化物歧化酶(SOD)、过氧化氢酶(CAT)等抗氧化酶活力及抗氧化相关基因表达的影响;用急性实验研究HFE对果蝇生存时间的影响。结果与对照组相比,1.5、4.5mg/ml HFE能够明显改善果蝇的抗氧化能力,果蝇体内SOD、CAT活力较空白对照组都有显著增加,丙二醛(MDA)含量显著下降;4.5mg/ml HFE组果蝇体内SOD、CAT m RNA表达水平显著上调,雄蝇methuselah(MTH)呈下调趋势;急性实验中HFE组果蝇的寿命明显延长。结论 HFE在体内具有较强的抗氧化活性,并可能通过上调抗氧化基因表达实现延长果蝇寿命的作用。Objective To determine the effect of hawthorn fruit extract(HFE) on lifespan and antioxidant function in Drosophila melanogaster. Methods 2d-old male and female Drosophila melanogaster were randomly divided into four groups and supplemented with different doses(0, 0.5, 1.5, 4.5 mg/ml) of HFE. The lifespan, activities of antioxidant enzymes and their encoding genes expressions were detected. Challenge tests were also carried out to investigate the effects of HFE on lifespan. Results Compared with the control, HFE at 1.5 and 4.5mg/ml could significantly improve antioxidant function, increase the activities of SOD and CAT and decrease MDA level. The expressions of SOD and CAT mRNA in Drosophila with 4.5mg/ml HFE were up-regulated, while the methuselah(MTH) mRNA expression was down-regulated. The lifespan of Drosophila treated with HFE was significantly extended in the challenge tests. Conclusion HFE possesses significant antioxidant capacity in vivo and can prolong the lifespan of Drosophila possibly via up-regulation of antioxidant genes expression.
