摘 要:目的:探讨大鼠氟乙酰胺中毒后心肌损伤情况及乙酰胺联合前列地尔注射液对心肌损伤的保护作用。方法:将100只Wistar大鼠随机分为正常对照组(A组),氟乙酰胺染毒组(B组),乙酰胺治疗组(C组),乙酰胺+前列地尔注射液治疗组(D组),前列地尔注射液治疗组(E组)。观察染毒后不同时间点(4h、1d、3d、7d)心肌组织形态及超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量变化。结果:氟乙酰胺中毒大鼠心肌细胞呈局灶性或片状变性、坏死,伴不同程度炎性细胞浸润;损伤于第1天出现,第3天最重,D组、C组第7天开始出现肉芽组织。与C组、E组比较,D组心肌损伤范围小,程度轻,心肌MDA含量明显降低(P<0.01);3组SOD活性比较差异无统计学意义。结论:氟乙酰胺可致心肌细胞局灶性或片状变性、坏死,肌纤维溶解。乙酰胺联合前列地尔注射液对氟乙酰胺中毒所致心肌损伤具有保护作用,其机制可能与乙酰胺阻断毒物体内进一步发展、前列地尔注射液抗氧化作用有关。Objective:The main purpose is to investigate the myocardial damage degree of rats and protective effect of acetamide combined with alprostadil injection on myocardium.Method:100 Wistar rats were randomly divided into normal control group(group A),fluoroacetamide poisoning group(group B),acetamide treatment group(group C),acetamide+ Alprostadil Injection treatment group(group D),Alprostadil Injection treatment group(group E).The changes of cardiomyocytes and the content of SOD and MDA were observed at different time points(4h,1d,3d,7d).Result:The myocardium of fluoroacetamide poisoning rats appeared focal or large-scale necrosis,with different degrees of inflammatory cells infiltrating.Myocardial injury appeared in 1dand developed into the most severe stage in 3d.Myocardial injury started to appear the granulation organization in group C and group D in 7when group D was compared with group C and group E,and the myocardial injury extent is smaller and lesser.The content of MDA in group D was obviously lower than that in group C and E(P〈0.01);The content of SOD in three groups had no statistical significance.Conclusion:Fluoroacetamide poisoning can lead to focal or large-scale necrosis of myocardium,with the infiltration of inflammatory cells.Acetamide combined with Alprostadil Injection have a protective effect on myocardial injury of the rats of fluoroacetamide poisoning.The possible mechanism is that acetamide block fluoroacetamide for further development.Alprostadil Injection has an effect of antioxygenation.
