摘  要:目的探讨甘草酸二铵(DG)对百草枯(PQ)中毒致急性肾损伤大鼠的保护作用及其可能机制。方法将50只SD大鼠按随机数字表法分为模型组(PQ组,n=20)、空白对照组(NS组,n=10)和干预组(DG组,n=20),PQ组和DG组大鼠给予PQ 100mg/kg一次性灌胃,NS组大鼠则给予等量生理盐水灌胃。DG组大鼠灌胃后立即给予甘草酸二铵50mg/kg于腹腔注射,每24h重复给药1次,直至72h后处死大鼠。取大鼠腹主动脉血,评估肾功能;病理学观察大鼠肾组织形态学改变情况,ELISA检测血清和肾组织IL-17及IL-16的含量。结果 HE染色结果显示,NS组大鼠肾组织结构正常;PQ染毒组大鼠可见肾小管管腔狭窄,甚至管腔闭锁,间质充血,细胞核固缩,细胞结构消失,肾小球结构紊乱;DG干预组大鼠肾组织病理改变较PQ染毒组大鼠明显减轻。肾功能结果显示,DG组大鼠血清Cr、BUN的水平明显低于PQ组(P<0.01),但显著高于NS组(P<0.01)。Elisa结果显示,DG组大鼠血清和肾组织中IL-6和IL-17的含量明显低于PQ组(P<0.05),但显著高于NS组(P<0.05)。结论甘草酸二铵可以减轻百草枯中毒大鼠的急性肾损伤,其机制可能与降低血清和肾组织中IL-6和IL-17的表达水平相关。Objective To investigate the protective effects and mechanisms of diammonium glycyrrhizinate(DG)on rats with acute renal injury induced by paraquat(PQ)poisoning. Methods Fifty SD rats were divided into three groups by table of random number:paraquat group(PQ group),glycyrrhizin diammonium intervention group(DG group)and control group(NS group).And there were 20 rats in PQ group,20 in DG group and 10 in NS group.The paraquats(100mg/kg)were administered by gavage in the PQ group and the DG group,while the NS group with the same dose of saline.Then,immediately,the glycyrrhizin diammonium[50mg/(kg·24h)]were administered by intraperitoneal injection in the DG group,while the same dose of saline were injected in the PQ group and NS group.The rats which were observed to 72 hours after administration were sacrificed.The blood of abdominal aortic were used to assess renal function.Morphological changes of renal tissue were observed by pathologica way.The ELISA assay was applyed to measure the IL-17 and IL-16 of kidneys and blood. Results The pathomorphological observation showed that the renal organizational structure was clear and there were no significant pathological changes in the NS group.While luminal stenosis and atresia,congestion of intercellular substance,damage of cell structure and disorder of glomerular structure were observed in the PQ group,and lesions in the DG group were lighter than the PQ group.The results of renal function test confirmed the levels of serum Cr,BUN of the DG group were lower than those in the PQ group(P〈0.01),while they were higher than the NS group(P〈0.01).The results of Elisa demonstrated that IL-6and IL-17 in serum and kidney of the DG group were less than those of the PQ group(P〈0.05),while they were more than the NS group(P〈0.05). Conclusion The diammonium glycyrrhizinate can alleviate acute renal injury in rats with paraquat poisoning.The mechanisms may be associated with the decrease of expressions of IL-6and IL-17 in serum a
