摘 要:目的:探讨细胞分裂周期蛋白6(CDC6)和同源盒异型基因5(HOXA5)在食管鳞状细胞癌组织中的表达及临床意义。方法采用免疫组织化学 SP 法检测51例食管鳞状细胞癌组织和27例正常食管黏膜组织中 CDC6和 HOXA5蛋白的表达情况。分析 CDC6及 HOXA5蛋白的表达与食管鳞状细胞癌临床病理特征的关系及食管鳞状细胞癌中 CDC6与 HOXA5蛋白的相关性。结果 CDC6和HOXA5在食管鳞状细胞癌组织中的阳性表达率分别为66.7%(34/51)、60.8%(31/51),均明显高于正常食管黏膜组织中的阳性表达率18.5%(5/27)、22.2%(6/27)。两组 CDC6及 HOXA5的阳性表达差异有统计学意义(χ^2=16.370,P =0.000;χ^2=10.528,P =0.001);CDC6和 HOXA5蛋白在食管鳞状细胞癌中的表达与患者细胞分化程度(χ^2=9.031,P =0.011;χ^2=10.064,P =0.007)和肿瘤 TNM 分期(χ^2=10.474,P =0.015;χ^2=11.667,P =0.009)有相关性,与患者的年龄(χ^2=0.000,P =1.000;χ^2=0.001, P =0.972)、性别(χ^2=0.000,P =1.000;χ^2=0.107,P =0.743)及淋巴结转移(χ^2=3.186,P =0.074;χ^2=2.212,P =0.137)无相关性;两种蛋白在食管鳞状细胞癌中表达呈正相关(r =0.454,P =0.001)。结论CDC6和 HOXA5蛋白在食管鳞状细胞癌中的高表达均明显高于食管正常组织,且与肿瘤的 TNM 分期及组织分化程度密切相关,提示 CDC6及 HOXA5的高表达与食管鳞状细胞癌的发生、发展以及增殖密切相关。Objective To investigate the expression and clinical significance of cell division cycle 6 (CDC6)and homeobox gene A5(HOXA5)in esophageal squamous cell carcinoma. Methods The expres-sion of CDC6 and HOXA5 in 51 specimens esophageal squamous cell carcinoma and 27 normal specimens esophageal tissues were evaluated by immunohistochemistry. Analyzed the relationship among the expression of CDC6 and HOXA5 protein and the clinicopathologic features of esophageal squamous cell carcinoma,along with the correlation between these two proteins. Results Immunohistochemical results showed that the positive expression rate of CDC6 and HOXA5 in esophageal squamous cell carcinoma tissue were 66. 7%(34 / 51)and 60. 8%(31 / 51),respectively,significantly higher than those in normal esophageal tissue18. 5%(5 / 27), 22. 2%(6 / 27),(χ^2 = 16. 370,P = 0. 000;χ^2 = 10. 528,P = 0. 001);There were significant positive correlation in the expression of CDC6 and HOXA5 and histological type(χ^2 = 9. 031,P = 0. 011;χ^2 = 7. 372,P = 0. 000), TNM stage(χ^2 = 10. 474,P = 0. 015;χ^2 = 11. 667,P = 0. 009),and there were no correlation in the expression of CDC6 and HOXA5 and age(χ^2 = 0. 000,P = 1. 000;χ^2 = 0. 001,P = 0. 972),sex(χ^2 = 0. 049,P = 0. 824;χ^2 = 0. 107,P = 0. 743),lymph node metastasis(χ^2 = 3. 186,P = 0. 074;χ^2 = 2. 212,P = 0. 137)in esophageal squamous cell carcinoma tissues. The expression of CDC6 and HOXA5 showed a positive correlation( r =0. 454,P = 0. 001). Conclusion The positive expression rate of CDC6 and HOXA5 in esophageal squamous cell carcinoma tissue were significantly higher than in normal esophageal tissue and close correlation with TNM stage and differentiation. High expression of CDC6 and HOXA5 may play important roles in the occurrence, development and proliferation of esophageal squamous cell carcinoma.
