蜂毒注射液对前列腺癌细胞凋亡的影响研究

摘　　要：目的研究蜂毒注射液(FD)对前列腺癌细胞凋亡的影响及可能机制。方法实验设立空白对照组和蜂毒注射液不同浓度组,用MTT方法检测前列腺癌细胞活性,Annexin V/PI染色流式细胞术检测细胞凋亡,同时用流式细胞术检测细胞内Ca2+浓度。结果与对照组比较,作用24小时、48小时、72小时后,FD8μg/ml,12μg/ml,16μg/ml,20μg/ml,24μg/ml组活性降低,8μg/ml作用24小时后细胞凋亡增多(P<0.05),4μg/ml,8μg/ml作用24小时,细胞内Ca2+浓度增高。结论蜂毒注射液可以诱导前列腺细胞凋亡,其作用可能与细胞内Ca2+超载相关。Objectives To study the effect of Apistoxin Injection（FD） on apoptosis of prostate cancer PC-3 cell, and to investigate the potential mechanism. Methods Blank control Group and FD groups of different concentrations were set up in this study. Cell proliferation was analyzed by MTT assay. Cell apoptosis was measured by Annexin V/PI staining and Flow Cytometry, The intracellular concentration of Ca2＋ measured by Flow Cytometry. Results Compared with the control, after treatment by FD for 24h,48h and 72h, cell activity decreased in FD groups of 8μg/ml,12μg/ml,16p, g/ml,20p, g/ml,24μg/ml. After treatment by FD for 24h, PC-3 cell of group 8p, g/ml apoptosis increased（ P 〈 0. 05 ）, the intracellular concentration of Ca2＋ in group 4μg/ml, 8μg/ml increased. Conclusions FD can induce PC-3 cell apoptosis, which may due to intraeellular Ca2＋ overload.
