摘 要:目的探讨外源性导入肝细胞生长因子(HGF)对人肺腺癌A549细胞增殖、黏附、移动和侵袭能力的影响及对其受体c-Met和上皮钙粘蛋白(E-cad)表达的调节作用。方法培养人肺腺癌细胞系A549细胞,按照给予外源性HGF(rh HGF)的浓度分为4组(0、20、40、80 ng/m L)。运用MTT法检测HGF对细胞增殖、黏附的影响,运用transwell小室观察各组间A549细胞侵袭和转移的变化,并采用免疫细胞化学检测rh HGF对c-Met和E-cad的作用。结果 rh HGF明显促进A549细胞增殖,提高黏附能力,40 ng/m L组最显著。rh HGF处理组A549细胞移动、侵袭能力明显增加(P<0.05)。rh HGF刺激下,cMet表达无明显改变,E-cad表达下调。结论 HGF可通过下调E-cad的表达来增强肿瘤细胞移动、侵袭能力,促进NSCLC的恶性进展。Objective To investigate the influence of exogenous hepatocyte growth factor (HGF) on proliferation, adhesion, migration and invasion in lung adenocarcinoma A549 cell and to detect the expression of c-Met and E-cadhrein (E-cad) proteins regulated by HGF. Methods Human lung adenocarcinoma A549 cell was cultivated, cell line was divided into four groups (0, 20, 40, 80 ng/mL) according to the concentrations of exogenous HGF-human recombinant HGF (rhHGF). MTT method was used to detect the effect of HGF on proliferation and adhesion. The difference of migration and invasion in four groups was observed by transwell model. The expressions of c-Met and E-cad proteins were detected by immunocytochemistry in the A549 cells treated by rhHGF. Results rhHGF significantly promoted proliferation and adhesion of A549 cell, and the most significant change was in 40 ng/mL group. The migration and invasion of A549 cell significantly increased after treated by rhHGF (P 〈 0.05). After stimulated by rhHGF, E-cad ex- pression decreased, but c-met expression had no change. Conclusion HGF can improve migration and invasion of tumor cell, promote NSCLC malignant progression by down-regulation of E-cad protein expression.
