摘 要:目的 探索不同年龄女性脂肪干细胞在细胞形态、表面标志物表达、生长特性及分化潜能方面的差异.方法 获取不同年龄女性脂肪组织,分离出脂肪干细胞,观察细胞形态,比较细胞增殖能力;采用流式细胞术检测细胞的免疫表型,诱导分化检测成脂、成骨分化潜能.结果 (1)所有脂肪干细胞均呈纤维细胞样;(2)随着年龄的增长,脂肪干细胞的增殖能力呈下降趋势;(3)P0代时,所有细胞均表达CD29、CD31、CD34、CD45和CD90,且随着年龄增加,表达呈下降趋势.传至P4代,CD34、CD45逐渐消失,CD31表达显著降低,CD29、CD90表达高于95%;(4)随着年龄的增长,脂肪干细胞的成脂、成骨分化潜能均呈下降趋势.结论 低年龄的脂肪干细胞可能具备更好的临床应用潜能.Objective To explore the differences of the morphology, surface marker expression, growth characteristics, and differentiation potential of adipose stem cells obtained from females at different ages ( 19 ~ 48 years old). Methods The adipose tissues from females at different ages were harvested and adipose-de- rived stem cells were separated. Cell morphology was observed and cell proliferation was compared. Cell immu- nophenotypes were detected using flow cytometry. Adipogenic and osteogenic differentiation were induced. Re- suits (1) All adipose-derived stem ceils were fibroblast-like. (2) The proliferation capacity of the adipose-de- rived stem cells was decreased with the increasing of the age. (3) The expression of CD29, CD31, CD34, CD45 and CD90 in all cells at passage 0 (P0) were positive and decreased with the increasing of the age. At Pd, the expressions of CD34 and CD45 were gradually disappeared, with CD31 significantly decreased, while the CD29 and CD90 expressions were higher than 95%, respectively. (4) With the increasing of the age, their adipogenic and osteogenic differentiation potentials were decreased. Conclusion Adipose-derived stem ceils from female at low age show better potentials for clinical application.
