摘　　要：目的:探讨ZEB-1对非小细胞肺癌细胞株A549生物学行为的影响。方法:构建干扰ZEB-1的质粒,转染至A548细胞株,运用体外增殖、迁移、侵袭实验来观察ZEB-1表达下调后对非小细胞肺癌细胞株A549生物学行为的影响。结果:下调ZEB-1的表达可降低A549细胞株的增殖、迁移和侵袭能力。结论:ZEB-1表达水平与非小细胞肺癌侵袭转移的能力显著相关。Objective：To study the effect of ZEB-1 on the behaviour of the non-small-cell lung cancer. Meth-ods：The exPression of ZEB-1 was down-regulated by RNA interference（ RNAi）in lung cancer cell line A549 and identify by Realtime-PCR and Western blot. In colony-forming assay,viro scratch test,Transwell based methods were used to detect the effectiveness of ZEB-1 on the Proliferation,migration and invasion ability of non-small cell lung cancer cells. Results：The successful transfection was confirmed by Realtime-PCR and Western blot in the cell line of A549. Down regulation of ZEB-1 can decrease Proliferation,migration and invasion Potential in A549 cell line. Conclusion：The exPression of ZEB-1 is significantly correlated with invasion and metastasis Potential in non-small-cell lung cancer.
