摘 要:目的:检测肺癌组织及癌旁正常组织中miR-34b和miR-155的表达水平及其与肺癌临床病理特征之间的关系。方法:收集50例非小细胞肺癌患者的肺癌组织和癌旁正常肺组织标本,运用Real-time PCR检测miR-34b和miR-155在肺癌组织和癌旁正常肺组织中的表达情况,分析其表达与肺癌临床病理特征之间的关系。结果:Real-time PCR检测显示在50例非小细胞肺癌组织中miR-34b的表达显著低于癌旁正常肺组织(P=0.019),而miR-155的表达显著高于癌旁正常肺组织(P=0.000)。MiR-34b和miR-155的表达与淋巴结转移情况显著相关(P均<0.05,miR-34b呈负相关,miR-155呈正相关);与性别、年龄、病理类型及肿瘤分化程度无明显相关;miR-34b的表达与临床病理分期无显著相关,而miR-155表达与临床病理分期显著相关。结论:MiR-34b和miR-155的异常表达与非小细胞肺癌的发生发展相关,其可能成为肺癌的筛查、诊断及治疗的肿瘤标志物。Objective:To investigate the expression of miR-34b and miR-155 in lung cancer tissue and adjacent normal tissue,and its relationship with chmicopathological features of lung cancer.Methods:The specimens of lung tumor tissue and adjacent normal lung tissue of 50 cases of lung cancer received surgical treatment in our department were collected.The expressions of miR-34b and miR-155 was detected by Real-time PCR.The correlations between the expression of miR-34b and miR-155 and clinicopathologic features were analyzed.Results:The expression of miR-34b in lung cancer tissue was lower than adjacent normal tissue (P =0.019),and the expression of miR-155 in lung cancer was higher than adjacent normal tissue(P =0.000).Significant negative correlation were observed between miR-34b expression levels and lymph node metastasis(P < 0.05),and significant positive correlation was observed between miR-155 expression levels and lymph node metastasis(P <0.05).It had no relationship with gender,age,pathological types and cell differentiation grade.Moreover,the expression of miR-155 was closely related to p-TNM stage,the expression of miR-34 b was not correlated to the type.Conclusion:The abnormal expression levels of miR-34b and miR-155 in NSCLC tissue indicates that it may play a role in the progression of NSCLC,and may be a useful molecular marker for screening,diagnosis and treatment of NSCLC.
