摘  要:目的探讨参芎化瘀胶囊对脑缺血-再灌注损伤的保护作用及机制。方法通过改良的Pulsineli 4血管阻断(4-VO)法制作全脑缺血-再灌注大鼠模型。72只雄性SD大鼠随机分成假手术组、模型组、参芎化瘀胶囊组。术后首先应用尼氏体和TUNEL染色研究缺血-再灌注后大鼠海马区神经元的形态学变化和凋亡情况,然后以免疫组织化学法检测海马区p38MARK的表达。结果假手术组相比,模型组可见大量的凋亡细胞,p38MARK蛋白表达增多,参芎化瘀胶囊组的凋亡细胞数量较模型组明显减少(p<0.05),p38MARK蛋白表达水平较低(p<0.05)。结论参芎化瘀胶囊对脑缺血-再灌注损伤的保护作用与调控p38MAPK信号通路抑制神经细胞凋亡有关。ObjectiveToexploretheprotectivepathogenesisandeffectofShenxionghuayucapsuleabout cerebral ischemia reperfusion injury .Methods The model of global cerebral ischemia reperfusion in rats was maked by improved four-vessel occlusion as described by pulsinelli .72 male SD rats were randomly divided into sham operation group , model group , Shenxionghuayu capsule group .After the first application of Nissl and TUNEL staining ischemic-morphological changes and apoptosis of neurons in the hippocampus of rats after reperfusion , then immunohistochemistry to detect the expression of hippocampus p 38MARK.Results Compared with the sham operation group, model group showed a large number of apoptotic cells , the expression of p38MARK protein increased, Shenxionghuayu capsule group , the number of apoptotic cells was significantly decreased than that in model group (p〈0.05), the expression of p38MARK protein was lower( p〈0.05).Concluis on The protective effect of Shenxionghuayu capsule on cerebral ischemia reperfusion injury may be p 38MAPK signaling pathway in protection of inhibits neural cells apoptosis .
