摘 要:目的:系统评价中国人群X线修复交叉互补基因1(X-ray repair cross complementing group 1,XRCC1)Arg399Gln基因多态性与肝癌易感性的关系.方法:在Pub Med、MEDLINE、EMBASE、CNKI、CBM、VIP及万方数据库中检索2000-01-01/2015-01-10发表的所有有关中国人群XRCC1 Arg399Gln基因多态性与肝癌易感性关系的相关文献.按照纳入和排除标准独立选择文献、提取资料,采用Stata12.0软件进行Meta分析,计算合并比值比(odds ratio,OR)及其95%可信区间(95%confidence interval,95%CI),并进行敏感性分析和发表偏倚的估计.结果:按照入选标准,共纳入13个研究,包括2972例患者和3789例对照者.Meta分析结果显示,与基因型Arg/Arg、Arg/Arg+Arg/Gln分别进行比较,基因型Gln/Gln均增加中国人群罹患肝癌风险(OR=1.47,95%CI:1.24-1.74,P<0.001;O R=1.26,95%C I:1.08-1.48,P=0.003);与基因型Arg/Arg进行比较,基因型Gln/Gln+Arg/Gln增加中国人群罹患肝癌风险(OR=1.49,95%CI:1.21-1.83,P<0.001).与等位基因Argalele比较,等位基因Gln-allele增加中国人群罹患肝癌的风险(OR=1.33,95%CI:1.16-1.54P<0.001).结论:XRCC1 Arg399Gln基因多态性与中国人群肝癌易感性相关,基因型Gln/Gln增加中国人群罹患肝癌的风险.AIM:To explore the correlation between X-ray repair cross complementing group 1(XRCC1)Arg399Gln polymorphism and susceptibility to hepatocellular carcinoma in Chinese populations.METHODS:All studies related to XRCC1Arg399 Gln polymorphism and hepatocellular carcinoma risk in Chinese subjects published from January 1,2000 to January 10,2015 were searched in PubMed,MEDLINE,EMBASE,CNKI,CBM,VIP and WanFang databases.We identified the studies according to inclusion and exclusion criteria.The pooled odds ratios(ORs) with 95%confidence intervals(CIs)were calculated using Stata 12.0 software.Publication bias and sensitivity were also evaluated.RESULTS:A total of 13 studies comprising2972 cases and 3789 controls were finally included.Meta-analysis showed that,compared to Arg/Arg and Arg/Arg +Arg/Gln,XRCC1 399Gln/Gln genotype increased the susceptibility to hepatocellular carcinoma in Chinese populations(OR = 1.47,95%CI:1.24-1.74,P 〈 0.001;OR = 1.26,95%CI:1.08-1.48,P = 0.003).Compared to Arg/Arg,Gln/Gln + Arg/Gln genotype increased the susceptibility to hepatocellular carcinoma in Chinese populations(OR = 1.49,95%CI:1.21-1.83,P 〈 0.001).Gln genotype increased the risk of hepatocellular carcinoma in Chinese populations(OR = 1.33,95%CI:1.16-1.54,P 〈0.001).CONCLUSION:XRCC1 Arg399 Gln polymorphism seems to be involved in hepatocellular carcinoma risk.XRCC1 399Gln/Gln genotype increases the susceptibility to hepatocellular carcinoma in Chinese populations.
