摘 要:目的 观察微小RNA (miRNA)-203在骨肉瘤中表达及其作用.方法 采用反转录-聚合酶链反应(RT-PCR)检测35例骨肉瘤患者癌及癌旁组织miRNA-203表达水平,同时检测骨肉瘤细胞株(MG-63、HOS、U2OS)与正常成骨细胞株hFOB1.19的miRNA-203表达.通过噻唑蓝比色法(MTT)与Transwell实验观察miRNA-203对MG-63增殖、侵袭的影响.结果 相对于癌旁组织与hFOB1.19细胞,miRNA-203在骨肉瘤癌组织与骨肉瘤细胞株表达显著下调(P<0.01).在MG-63细胞株过表达miRNA-203可显著抑制细胞增殖(P<0.01),48、72 h的抑制率分别为21.32%、30.20%,miRNA-203组侵袭细胞数[(50±8)个]显著低于空载体组的(131±5)个(P<0.05).结论 miRNA-203在骨肉瘤中表达明显降低.Objective To explore the expression and role of microRNA (miRNA)-203 in osteosarcoma.Methods Reverse transcriptase-polymerase chain reaction (RT-PCR) assays were used to detect the expression of miRNA-203 in human osteosarcoma and adjacent normal tissues.The expression of miRNA-203 in osteosarcoma cell lines (MG-63,HOS,U2OS) and normal osteoblast cell hFOB1.19 was also tested.Methyl thiazol tetrazolium (MTT) and Transwell assays were applied to observe the role of miRNA-203 in proliferation and invasion of MG-63.Results miRNA-203 was significantly down-regulated in human osteosarcoma tissues and MG-63 as compared with adjacent normal tissues and hFOB 1.19 (P < 0.01).Overexpression of miRNA-203 inhibited MG-63 proliferation (P < 0.01).At the time points of 48 and 72 h,the inhibition rate was 21.32%,and 30.20% respectively.Morever,the invasion cells of miRNA-203 overepression (50 ± 8) were significantly less than those of control group (131 ± 5,P < 0.05).Conclusion miRNA-203 may play an important role in the osteosarcoma progression.
