摘 要:目的 观察Endocan在胶质瘤中的表达,探讨其与血管内皮生长因子(VEGF)、微血管密度(MVD)和细胞核增殖抗原(Ki-67)的相互关系及其临床意义.方法 应用免疫组织化学的方法分析183例胶质瘤和32例正常脑组织中Endocan、VEGF、MVD、Ki-67的表达.同时收集51例新鲜胶质瘤标本,采用RT-PCR方法检测Endocan mRNA的表达水平.比较Endocan、VEGF与胶质瘤病理级别、MVD、Ki-67的关系及其表达的相关性.结果 (1)Endocan、VEGF的表达水平随病理级别的增高而增多(P <0.01);(2) Endocan、VEGF的表达与CD34标染的MVD呈显著正相关(r=0.55、0.64,P<0.01);(3)Endocan、VEGF的表达与Ki-67呈显著正相关(r=0.43、0.58,P<0.01);(4)在低级别(WHO Ⅰ、Ⅱ级)和高级别(WHO Ⅲ、Ⅳ级)胶质瘤中,Endocan mRNA表达水平的差异有统计学意义(P<0.01).结论 胶质瘤中Endocan表达水平越高,则胶质瘤的恶性程度、MVD及增殖活性越高.Objective To investigate the expression and clinical significances of endocan,vascular endothelial growth factor (VEGF) and CD-34 in hunman brain gliomas.Methods We investigate the expression levels of endocan mRNA with reverse transcription-polymerase chain reaction (RT-PCR) in 51 cases gliomas.Immunohistochemistry was used to detect the expression levels of endocan,VEGF,CD-34 and proliferation cell nuclear antigen (Ki-67) in 32 cases normal brain and 183 cases gliomas.Their correlation to the pathologic grade,and the relationship between the expression of endocan and micro vascular density (MVD) in bunman gliomas were studied.Results (1) The increasing expression levels of endocan and VEGF had positive correlation with the ascending of pathologic grade of tumor specimen (rcndocan =0.69,P <0.01;rVEGF =0.78,P <0.01).(2) There was a positive and significant correlation between the expression levels of MVD and endocan aswell as VEGF (r =0.55,P <0.01;r =0.64,P < 0.01).(3) There were positive and significant correlations between the expression levels of Ki-67 and endocan as well as VEGF (r =0.43,P < 0.01;r =0.58,P < 0.01).(4) The expression levels of endocan mRNA were significantly higher in high grade gliomas (WHO Ⅲ,Ⅳ) than those in low grade gliomas (WHO Ⅰ,Ⅱ) (P < 0.01).Conclusion The expression levels of endocan and VEGF play an important role in predicting the malignancy and proliferation of human gliomas.
