中药制剂联合美沙拉嗪对DSS诱导的小鼠结肠炎的治疗作用

摘　　要：目的探讨中药制剂联合美沙拉嗪对DSS诱导的小鼠结肠炎的治疗作用。方法以C57BL/6小鼠为研究对象,采用DSS饮水法建立结肠炎模型,随机分为4组:模型组、中药制剂组、美沙拉嗪组、美沙拉嗪+中药制剂组,分别于第4天给予生理盐水、中药制剂、美沙拉嗪、美沙拉嗪联合中药制剂灌胃。每日观察各组小鼠精神状态、大便性状及体重变化;于第10天收集小鼠粪便行隐血测定,计算疾病活动指数(DAI);断颈处死各组小鼠,取结肠炎病变严重部位测定髓过氧化物酶活力(MPO);取结肠炎病变严重部位做病理切片,行HE染色评价病变严重程度。结果成功建立小鼠结肠炎模型,各治疗组的DAI、MPO和组织学评分均较模型组明显改善,其中联合治疗组较美沙拉嗪组明显改善,差异有统计学意义。结论复方中药制剂联合美沙拉嗪治疗不仅能改善DSS诱导的小鼠结肠炎的症状评分,也能改善其病理评分。Objective To investigate the therapeutic effect of traditional Chinese medicine（ TCM） combined with mesalazine on DSS-induced colitis in mice. Methods C57 BL /6 mice were used as research subjects and were given water containing dextran sulfate sodium（ DSS） to establish the model of colitis. They were randomly divided into four groups,mice in model group,TCM group,mesalazine group,TCM combined mesalazine group（ combined group）were given normal saline,TCM,mesalazine,TCM combined with mesalazine by gavage administration from the fourth day respectively. The mental state,defecate traits and changes of body weight were observed every day. The disease activity index（ DAI） was calculated. All the mice were sacrificed on the 10 th day,and the inflammation degree of colon was assessed by HE dyeing as well as myeloperoxidase（ M PO） activity assay. Results Colitis models of mice were established successfully. Compared with model group,DAI,M PO activity,microscopic inflammation score in treatment groups were improved significantly,and the levels in combined group were lower than those of mesalazine group. Conclusion The combined treatment of traditional Chinese medicine and mesalazine can improve not only the symptom score,but also the microscopic inflammation score in colitis mice induced by DSS.
