摘  要:目的观察S-亚硝基谷胱甘肽(GSNO)对人肺腺癌A549细胞的作用,为GSNO作为药物来治疗肺癌提供依据。方法培养的A549细胞分别给予0.5、1.0、1.5、2.0、2.5 mmol/L GSNO 5个剂量,并分别培养24、48、72 h,应用CCK8试剂检测IC50;应用CCK8试剂检测二硫苏糖醇(DTT)逆转GSNO作用的给药时间点;应用原位凋亡荧光检测试剂盒检测GSNO给药组和GSNO联合DTT给药组中A549细胞的凋亡情况。结果 GSNO分别处理24、48、72 h后检测,结果 IC50分别为18.174、2.020、1.836 mmol/L;在GSNO给予20、40 min后给予DTT,结果 OD值显著提高(P<0.05),因而确定DTT给药时间点为GSNO给药后20 min;GSNO给药组显示凋亡细胞大量增加,GSNO联合DTT给药组则逆转了A549细胞的大量凋亡。结论 A549细胞凋亡可能是GSNO抑制细胞生长的一个重要机制,GSNO诱导的A549细胞凋亡可能与巯基-亚硝基化修饰作用有关。GSNO能促使A549细胞凋亡,该过程对于人肺腺癌的治疗具有明显的指导意义。Objective To observe the effect of S-nitrosoglutathione(GSNO) on human lung adenocarcinoma A549 cells and to provide theoretical basis for the treatment of lung cancer by GSNO. Methods A549 cells were treated with0.5,1.0,1.5,2.0,2.5 mmol/L GSNO respectively for 24,48,72 h.CCK8 was applied to test IC50 of GSNO and dithiothreitol(DTT) administration time.Fluorescence situ apoptosis detection kit was to detected the apoptosis of A549 cells in GSNOtreated group and DTT +GSNO-treated group. Results The IC50 values were respectively 18.174,2.020,1.836 mmol/L after treatment of GSNO for 24,48,72 h.Treatment with DTT 20 and 40 min after administration of GSNO indicated significantly improved OD(P〈0.05),so 20 min was applied in the following test.GSNO-treated group revealed increased apoptotic A549 cells,reversed by co-treatment with DTT +GSNO-treated group. Conclusion Apoptosis of A549 cells may be an important mechanism for the inhibition of GSNO.Apoptosis of A549 cells induced by GSNO may be relative to S-nitrosylation.GSNO can lead to apoptosis of A549 cells,so this process has obvious guiding significance for the treatment of human lung adenocarcinoma.
