柱前衍生RP-HPLC法测定苍耳子和意大利苍耳子中氨基酸的种类

摘　　要：对苍耳子和意大利苍耳子中游离和水解氨基酸种类及提取液中各氨基酸质量的测定，建立一种柱前衍生 RP-HPLC 法。用50％乙醇作溶剂超声提取苍耳子和意大利苍耳子中的游离氨基酸，用6 mol·L－1 HCl 水解提取出其水解氨基酸，柱前衍生化试剂选用异硫氰酸苯酯（PITC）。测定出苍耳子和意大利苍耳子中含20种氨基酸，且其在0．0391～1．250 mmol·L－1均呈现良好的线性关系（R2均大于0．9993），平均回收率为92．70％～98．44％，相对标准偏差值为0．70％～3．27％。该方法具有良好的灵敏度、准确性、重复性和稳定性，分离出了苍耳子和意大利苍耳子中的20种氨基酸，并计算出其提取质量。In this study,the types and content of amino acids in Xanthium sibiricum Patr.（XSP）and Xanthiumitalicum Moretti （XIM）were determined,and a precolumn derivatization RP-HPLC method was established.The free amino acids in XSP and XIM were ultrasonically extracted by 50% ethanol as solvent.The hydrolyzed amino acids were extracted by hydrolysis method in HCl （6 mol·L-1 ）.Phenylisothiocyanate was selected as the precolumn derivatization agent to analyze the amino acids by gradi-ent elution.The results indicate that twenty kinds of amino acids in XSP and XIM are characterized and they have good linearity within the range of 0.039 1-1.250 mmol·L-1 （R2 ≥ 0.999 3）.The average recoveries are 92.70%-98.44% and the relative standard deviation （RSD）values are between 0.70% and 3.27%.The method has good sensitivity,accuracy,repeatability and stability.Twenty kinds of amino acids are isolated from the XSP and XIM,and the extraction quality is calculated.
