不同因素对吗啡诱导的小鼠CPP实验的影响

摘  要:为了比较不同因素对吗啡诱导的小鼠条件性位置偏爱(CPP)实验模型的影响,以建立理想的CPP模型,本实验首先筛选出符合条件的小鼠来注射吗啡(5 mg·kg-1),分为皮下注射和腹腔注射2种不同给药方式组,于不同给药时间组连续皮下注射吗啡3 d,5 d,7 d,并于不同测试时间(15 min或30 min)组,分别对小鼠进行CPP训练,然后进行测试,同时以小鼠在伴药箱(白箱)的活动时间为指标进行比较.结果表明:皮下注射和腹腔注射生理盐水组与吗啡组相比较,均存在极显著性差异(P<0.001);皮下注射吗啡组与腹腔注射吗啡组的差异性不显著(P>0.05),连续给药3 d,5 d,7 d均能成功建立CPP模型(P<0.001),CPP训练后,3 d吗啡组与5 d、7 d吗啡组相比较,均存在极显著差异(P<0.001),5 d与7 d吗啡组相比较,则存在显著性差异(P<0.05).在连续给药5 d的基础上,不同测试时间(15 min与30 min)吗啡组与生理盐水组相比较,小鼠在伴药箱的活动时间具有极显著差异性(P<0.001).由此得出结论:皮下注射和腹腔注射2种给药方式对吗啡诱导的小鼠CPP模型的建立无影响;连续注射5 d所建立的CPP模型更稳定,更经济,更理想;测试时间为15 min时更省时,更具代表性.To compare the effects of the different experimental conditions on morphine-induced conditioned place preference of mice and establish satisfactory CPP model,in the report,the eligible mice injected with morphine( 5mg /kg) were divided into different groups random for conditioned place preference( CPP) training according to intraperitoneal injection( i. p.) or subcutaneous injection( s. c.); 3 d,5 d,and 7 d different continuous injection time; and 15 min or 30 min different testing time. Each group mice received an injection of morphine( 5mg /kg) or saline,and then,the time spent in the drug-paired box was observed. The results indicated that it was obvious that morphine groups significantly induced CPP effect after training,but no CCP effects occurred in saline groups,whatever it was s. c. or i. p. injection( P 〈0. 001). CPP model could be constructed through3 d,5d,or 7d different continuous injection of morphine,the differences among these groups were significant.The time spent in drug-paired box was significant difference between 15 min and 30 min testing time based on 5d continuous injection of morphine( P 〈0. 01). These results suggested that the different injection s. c. or i. p.has no effects on morphine induced CPP model of mice; 5d continuous injection is the most stable,economical,and ideal condition for the construct of the morphine-induced CPP mice model; the test time of 15 min is the most time-saving and most representative.
